To explore the effect of high glucose on NADPH oxidase (NOX) expression and intracellular reactive oxygen species (ROS) generation in human umbilical vein endothelial cells (HUVECs).
HUVECs were divided into a control group, a mannite group, a glucose group and a glucose plus diphenylene iodonium (DPI) group. Intracellular ROS was detected by flow cytometry. RNA and protein expression of NOX in HUVECs was analyzed by reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot, respectively.
High glucose may increases intracellular ROS generation by increasing the expression of NOX4 in HUVECs, which might mediate the oxidative stress.
探讨高糖对人脐静脉内皮细胞(HUVECs)中NADPH氧化酶(NOX)表达及细胞内活性氧(ROS)生成的影响。
将HUVECs分为对照组、甘露醇组、葡萄糖组和葡萄糖加二苯基碘鎓(DPI)组。采用流式细胞术检测细胞内ROS。分别通过逆转录聚合酶链反应(RT-PCR)和蛋白质免疫印迹法分析HUVECs中NOX的RNA和蛋白质表达。
1)与对照组相比,葡萄糖组细胞内ROS显著增加(P<0.05,n=3),但葡萄糖加DPI组无显著变化(P>0.05,n=3);2)与对照组相比,葡萄糖组中NOX4的mRNA和蛋白质表达显著增加(P<0.05),但NOX2、p22phox、p67phox和rac的mRNA和蛋白质表达无显著变化(均P>0.05);3)葡萄糖加DPI组与对照组之间NOX2、NOX4、p22phox、p67phox和rac的mRNA和蛋白质表达无显著差异(均P>0.05)。
高糖可能通过增加HUVECs中NOX4的表达来增加细胞内ROS的生成,这可能介导氧化应激。
