Abstract

ETHNOPHARMACOLOGICAL RELEVANCE

Mahuang-Xixin-Fuzi Decoction (MXF) as a famous formula for the treatment of colds, fever, nasal congestion and headache with elder people, has always been widely used in traditional Chinese medicine. The present study is aimed at investigating the treatment effect of MXF on Kidney-Yang deficiency syndrome in mice simultaneously infected with H1N1 virus.

MATERIALS AND METHODS

We employed the Kidney-Yang deficiency mouse model to investigate the effect of MXF against influenza A virus (A/FM/1/47, H1N1). Mice were infected with the virus after fifteen days Kidney-Yang deficiency syndrome was established (intraperitoneal injection of estradiol benzoate), while MXF was orally administrated with 1.2-4.7g/kg/d for 6 consecutive days after inoculation. Body weight, rectal temperature, morbidity, and mortality were recorded daily. Histopathologic changes, antioxidant activity of SOD and MDA were detected. Moreover, levels of inflammatory cytokines including IL-6, IL-10, MCP-1, TNF-α were measured in the sera of mice.

RESULTS

We found that the extract of MXF at dosages of 2.3-4.7g/kg could effectively diminish mortality rate, ameliorate lung edema and inflammation. Administration of MXF decoction significantly depressed the expression of IL-6, MCP-1 and TNF-α, and markedly increased expression of IL-10 in serum. Simultaneously, the extract was also found to reduce MDA and increase SOD in the lung tissue of mice.

CONCLUSION

These data support the notion that the extract of MXF could treat Kidney-Yang deficiency syndrome in mice simultaneously infected with influenza A virus by reducing inflammation and increasing antioxidant activities.

麻黄-细辛-附子汤(MXF)作为一种治疗老年人感冒、发热、鼻塞和头痛的著名方剂,一直在传统中医中广泛应用。本研究旨在探讨MXF对同时感染H1N1病毒的小鼠肾阳虚证的治疗效果。

材料与方法

我们采用肾阳虚小鼠模型来研究MXF对甲型流感病毒(A/FM/1/47,H1N1)的作用。在建立肾阳虚证(腹腔注射苯甲酸雌二醇)15天后,小鼠感染病毒,而在接种后连续6天以1.2 - 4.7g/kg/d的剂量口服给予MXF。每天记录体重、直肠温度、发病率和死亡率。检测组织病理学变化、超氧化物歧化酶(SOD)和丙二醛(MDA)的抗氧化活性。此外,还测定了小鼠血清中包括白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、单核细胞趋化蛋白-1(MCP-1)、肿瘤坏死因子-α(TNF-α)在内的炎症细胞因子水平。

结果

我们发现,2.3 - 4.7g/kg剂量的MXF提取物可有效降低死亡率,改善肺水肿和炎症。给予MXF汤剂可显著降低血清中IL-6、MCP-1和TNF-α的表达,并显著增加IL-10的表达。同时,还发现该提取物可降低小鼠肺组织中的MDA含量并增加SOD含量。

结论

这些数据支持以下观点,即MXF提取物可通过减轻炎症和增强抗氧化活性来治疗同时感染甲型流感病毒的小鼠的肾阳虚证。
