Abstract

To investigate the effects of miR-382 on cell migration, invasion and proliferation of gastric cancer cell lines MGC-803. The level of miR-382 expression was detected by real-time RT-PCR in 50 paired gastric cancer tissues and their adjacent normal tissues. miR-382 overexpression was achieved by transfection of construct pcDNA-miR-382 into MGC-803 cells. The migration, invasion and proliferation of MGC-803 cells were detected by the scratch wound assay, Transwell and CCK-8, respectively. miR-382 was decreased in 41 cases (82%) of gastric cancer tissues compared to their control. Furthermore, overexpression of miR-382 effectively inhibited the migration, invasion and proliferation of MGC-803 cells(<0.05). Down-regulation of miR-382 has a correlation with the progression of gastric cancer. Up-regulation of miR-382 can inhibit the migration, invasion and proliferation of MGC-803 cells.

研究miR-382对胃癌细胞系MGC-803细胞迁移、侵袭及增殖的影响。采用实时逆转录聚合酶链反应(RT-PCR)检测50对胃癌组织及其癌旁正常组织中miR-382的表达水平。通过将构建体pcDNA-miR-382转染至MGC-803细胞实现miR-382的过表达。分别采用划痕实验、Transwell小室实验和CCK-8实验检测MGC-803细胞的迁移、侵袭及增殖能力。与对照相比,41例(82%)胃癌组织中miR-382表达降低。此外,miR-382过表达有效抑制了MGC-803细胞的迁移、侵袭及增殖(P<0.05)。miR-382下调与胃癌进展相关。上调miR-382可抑制MGC-803细胞的迁移、侵袭及增殖。
