Abstract

OBJECTIVE

The incidence of gastric cancer is very high all over the world, but the mechanism of the occurrence and development of gastric cancer is unclear. Secretory leukocyte protease inhibitor (SLPI) is overexpressed in gastric, lung and ovarian cancers, which accelerates the metastasis of cancer cells. In this research, we mainly explored the expression level and possible mechanism of SLPI in gastric cancer.

PATIENTS AND METHODS

The expression and clinical significance of SLPI in 68 cases of gastric cancer tissues and adjacent tissues were detected by qRT-PCR. Cell Counting Kit-8 (CCK8) assay was used to detect the proliferation ability of gastric cancer cell lines. In addition, we used Western blot to clarify the relationship between SLPI and metastasis.

RESULTS

Compared with the adjacent tissues, we found that SLPI was highly expressed in gastric cancer tissues. We also found that the expression of SLPI was in significant correlation with the survival time, clinical classification and size of the tumor. What's more, SLPI could promote the proliferation and metastasis of gastric cancer by regulating P53, Bcl-2 and Caspase-8 expression through apoptosis signaling pathway.

CONCLUSIONS

We concluded that SLPI was closely related with to invasion and metastasis of gastric cancer. Perhaps we can hopefully find new targets for the treatment of gastric cancer.

目的

胃癌在全球的发病率都很高,但其发生发展机制尚不清楚。分泌型白细胞蛋白酶抑制剂(SLPI)在胃癌、肺癌和卵巢癌中过表达,可加速癌细胞转移。本研究主要探讨SLPI在胃癌中的表达水平及可能机制。

患者与方法

采用qRT-PCR检测68例胃癌组织及癌旁组织中SLPI的表达及临床意义。采用细胞计数试剂盒-8(CCK8)法检测胃癌细胞系的增殖能力。此外,我们用蛋白质免疫印迹法阐明SLPI与转移之间的关系。

结果

与癌旁组织相比,我们发现SLPI在胃癌组织中高表达。我们还发现SLPI的表达与生存时间、临床分级及肿瘤大小显著相关。此外,SLPI可通过凋亡信号通路调节P53、Bcl-2和Caspase-8的表达,从而促进胃癌的增殖和转移。

结论

我们得出结论,SLPI与胃癌的侵袭和转移密切相关。或许有望找到治疗胃癌的新靶点。
