This study provides an in vivo evaluation of the inflammatory response, levels of cell proliferation and apoptosis, and the presence of necrosis after dental bleaching with two concentrations of hydrogen peroxide (HO).
Wistar rats were divided into Control (placebo gel), BLUE (20% HO, 1×50min), and MAXX (35% HO, 3×15min) groups. At 2 and 30days, the rats were killed (n=10). The jaws were processed for histology analysis and PCNA and Caspase-3-cleaved immunohistochemistry, and data were submitted to the Mann-Whitney or ANOVA test (P<0.05).
At 2days, the MAXX group showed necrosis and the BLUE group revealed moderate inflammation on the occlusal third of the crown (P<0.05). At 30days, tertiary dentin had formed and there was an absence of inflammation. The level of cell proliferation was higher in the middle third of the BLUE group (P<0.05), and cervical of MAXX at 2days (P<0.05), decreasing at 30days. The apoptosis was present at 2days, particularly in the cervical third of the crown in the bleached groups (P<0.05), with a decrease only at 30days in the BLUE group (P<0.05).
The concentration of HO influences effects on the pulp tissue, where a higher concentration of HO can cause necrosis in the pulp and a prolonged effect within the apoptotic process; lower concentrations of HO provide moderate inflammation, cell proliferation and apoptosis with a reduction of these processes over time.
本研究通过体内实验评价两种浓度(20%HO 和 35%HO)过氧化氢(HO)牙齿漂白后对炎症反应、细胞增殖和凋亡水平以及坏死的影响。
Wistar 大鼠分为对照组(安慰剂凝胶)、BLUE 组(20%HO,1×50min)和 MAXX 组(35%HO,3×15min)。第 2 天和第 30 天处死大鼠(每组 n=10)。对颌骨进行组织学分析和 PCNA 和 Caspase-3 切割免疫组化,数据采用 Mann-Whitney 或 ANOVA 检验(P<0.05)。
第 2 天,MAXX 组出现坏死,BLUE 组牙冠咬合三分之一处出现中度炎症(P<0.05)。第 30 天,形成第三期牙本质,无炎症。BLUE 组牙冠中部细胞增殖水平较高(P<0.05),MAXX 组牙颈在第 2 天(P<0.05)较高,第 30 天降低。第 2 天存在凋亡,尤其是在漂白组牙冠颈三分之一(P<0.05),第 30 天 BLUE 组仅减少(P<0.05)。
HO 浓度影响牙髓组织的影响,较高浓度的 HO 可导致牙髓坏死和凋亡过程延长;较低浓度的 HO 可引起适度的炎症、细胞增殖和凋亡,并随着时间的推移减少这些过程。
