Abstract

Porcine epidemic diarrhea virus (PEDV) causes acute diarrhea, vomiting, dehydration, weight loss, and high mortality rate in neonatal piglets. Porcine epidemic diarrhea (PED) has been reported in Europe, America, and Asia including Thailand. The disease causes substantial losses to the swine industry in many countries. Presently, there is no effective PEDV vaccine available. In this study, we developed a plant-produced monoclonal antibody (mAb) 2C10 as a prophylactic candidate to prevent the PEDV infection. Recently, plant expression systems have gained interest as an alternative for the production of antibodies because of many advantages, such as low production cost, lack of human and animal pathogen, large scalability, etc. The 2C10 mAb was transiently expressed in and lettuce using geminiviral vector. After purification by protein A affinity chromatography, the antibody was tested for the binding and neutralizing activity against PEDV. Our result showed that the plant produced 2C10 mAb can bind to the virus and also inhibit PEDV infection . These results show excellent potential for a plant-expressed 2C10 as a PEDV prophylaxis and a diagnostic for PEDV infection.

猪流行性腹泻病毒(PEDV)可导致新生仔猪出现急性腹泻、呕吐、脱水、体重减轻和高死亡率。欧洲、美洲以及包括泰国在内的亚洲均已报告发生猪流行性腹泻(PED)。该疾病在许多国家给养猪业造成了巨大损失。目前,尚无有效的PEDV疫苗。在本研究中,我们开发了一种植物生产的单克隆抗体(mAb)2C10作为预防PEDV感染的候选药物。近来,由于生产成本低、无人类和动物病原体、可大规模生产等诸多优势,植物表达系统作为生产抗体的一种替代方法受到了关注。2C10单克隆抗体利用双生病毒载体在烟草和生菜中瞬时表达。经蛋白A亲和层析纯化后,检测该抗体对PEDV的结合和中和活性。我们的结果表明,植物生产的2C10单克隆抗体能够与病毒结合,还能抑制PEDV感染。这些结果显示,植物表达的2C10作为PEDV预防药物和PEDV感染诊断试剂具有巨大潜力。
