Dendrobium officinale is one valuable traditional Chinese medicine, which has skyscraping medicinal value. Polysaccharide is the main active ingredient in D. officinale; its antioxidant activity is a hot research topic nowadays. Oxidative stress plays an important role in the pathological progress of a variety of cardiovascular disease, as one of key factors of cardiomyocyte apoptosis. This research adopts a model of HO induction-H9c2 cardiomyocytes apoptosis, aiming to study the effect of Dendrobium officinale Polysaccharide (DOP-GY) for cardiomyocyte apoptosis caused by oxidative stress and its possible mechanism. Our results showed that pretreatment of DOP-GY (low dose: 6.25μg/mL, medium dose: 12.5μg/mL, high dose: 25μg/mL) followed by a 2h incubation with 200μM HO elevated the survival rate, cutted the LDH leakage, reduced lipid peroxidation damage, improved the activity of the endogenous antioxidant enzymes. In addition, the pretreatment of DOP-GY significantly inhibited the production of ROS, declined of the mitochondrial membrane potential, down-regulated pro-apoptosis protein and up-regulated anti-apoptosis protein. The protective effect was correlated with the PI3K/Akt and MAPK signal pathway. Collectively, these observations suggest that DOY-GY has the potential to exert cardioprotective effects against HO-induced H9c2 cardiomyocyte apoptosis.
铁皮石斛是一种珍贵的传统中药,具有极高的药用价值。多糖是铁皮石斛的主要活性成分;其抗氧化活性是当今热门的研究课题。氧化应激在多种心血管疾病的病理进程中起重要作用,是心肌细胞凋亡的关键因素之一。本研究采用过氧化氢诱导H9c2心肌细胞凋亡模型,旨在探讨铁皮石斛多糖(DOP - GY)对氧化应激所致心肌细胞凋亡的影响及其可能机制。我们的结果表明,用DOP - GY(低剂量:6.25μg/mL,中剂量:12.5μg/mL,高剂量:25μg/mL)预处理,然后与200μM过氧化氢孵育2小时,可提高细胞存活率,减少乳酸脱氢酶泄漏,减轻脂质过氧化损伤,提高内源性抗氧化酶活性。此外,DOP - GY预处理显著抑制活性氧的产生,降低线粒体膜电位,下调促凋亡蛋白并上调抗凋亡蛋白。其保护作用与PI3K/Akt和MAPK信号通路相关。总体而言,这些观察结果表明DOP - GY对过氧化氢诱导的H9c2心肌细胞凋亡具有潜在的心脏保护作用。
