壳寡糖对人肝癌细胞的抑制作用

Abstract

BACKGROUND

Chitosan oligosaccharide, the degradation products of chitin, was reported to have a wide range of physiological functions and biological activities. In this study, we explored the inhibitory effect of Chitosan oligosaccharide on human hepatoma cells.

MATERIALS AND METHODS

MTT assay was applied to detect cell viability of the human hepatoma cells treated with Chitosan oligosaccharide. Flow cytometric analysis was used to investigate the apoptosis of the human hepatoma cells treated with Chitosan oligosaccharide. We employed western blot to investigate the underlying mechanisms involved in the apoptosis.

RESULTS

Our data indicated that chitosan oligosaccharide dose-dependently inhibited the growth of hepatoma cells and induced apoptosis. On the molecular level, chitosan oligosaccharide decreased Bcl-2 and increased Caspase-3 expression which may be related to the apoptosis of hepatoma cells.

CONCLUSION

Our results provide an experimental basis for the clinical development of Chitosan oligosaccharide as a novel anti-hepatoma drug.

背景

材料与方法

采用MTT法检测壳寡糖处理后人肝癌细胞的活力。运用流式细胞术分析壳寡糖处理后人肝癌细胞的凋亡情况。我们采用蛋白质免疫印迹法研究凋亡相关的潜在机制。

结果

我们的数据表明壳寡糖剂量依赖性地抑制肝癌细胞生长并诱导凋亡。在分子水平上,壳寡糖降低了Bcl-2表达并增加了Caspase-3表达,这可能与人肝癌细胞的凋亡有关。

结论

我们的结果为壳寡糖作为新型抗肝癌药物的临床开发提供了实验依据。
