Abstract

BACKGROUND/AIM: Medulloblastoma (MBL), an archetypal primitive neuroectodermal tumor of the cerebellum, is the most common pediatric central nervous system malignancy representing approximately 20% of all childhood brain tumors. Herein, we report on a new xenotransplantable tumor cell line, derived from a 6-year-old female patient with cerebellar medulloblastoma, and the completele proteome molecular characterization of subsequent tumors from MBL xenotrasplanted mice.

MATERIALS AND METHODS

Tumors were grown in nude mice as subcutaneous xenografts (MBLX) composed of small round cells with hyperchromatic nuclei and scant cytoplasm. Tumor specimen were extracted from animals upon their sacrifice and their molecular proteomic content was analyzed by 2-DE coupled to MALDI-TOF MS analysis.

RESULTS

Altogether 350 single-gene products were identified through the current approach, reported as the MBLX database.

CONCLUSION

This new xenotransplantable tumor model, offers the scientific community valuable insight on the validity of xenografts altogether, while providing the means for a novel experimental model towards the study of human MBL.

背景/目的:髓母细胞瘤(MBL)是小脑典型的原始神经外胚层肿瘤,是最常见的儿童中枢神经系统恶性肿瘤,约占所有儿童脑肿瘤的20%。在此,我们报告一种新的可异种移植的肿瘤细胞系,它源自一名6岁患小脑髓母细胞瘤的女性患者,以及对MBL异种移植小鼠后续肿瘤的完整蛋白质组分子特征分析。

材料与方法

肿瘤在裸鼠体内作为皮下异种移植物(MBLX)生长,由核深染、胞质稀少的小圆形细胞组成。在动物处死后提取肿瘤标本,并通过二维电泳结合基质辅助激光解吸电离飞行时间质谱分析对其分子蛋白质组含量进行分析。

结果

通过当前方法共鉴定出350种单基因产物,作为MBLX数据库公布。

结论

这种新的可异种移植肿瘤模型,为科学界提供了关于异种移植有效性的宝贵见解,同时为研究人类MBL提供了一种新的实验模型。
