Cancer stem cells can generate tumors from only a small number of cells, whereas differentiated cancer cells cannot. The prominent feature of cancer stem cells is its ability to self-renew and differentiate into multiple types of cancer cells. Cancer stem cells have several distinct tumorigenic abilities, including stem cell signal transduction, tumorigenicity, metastasis, and resistance to anticancer drugs, which are regulated by genetic or epigenetic changes. Like normal adult stem cells involved in various developmental processes and tissue homeostasis, cancer stem cells maintain their self-renewal capacity by activating multiple stem cell signaling pathways and inhibiting differentiation signaling pathways during cancer initiation and progression. Recently, many studies have focused on targeting cancer stem cells to eradicate malignancies by regulating stem cell signaling pathways, and products of some of these strategies are in preclinical and clinical trials. In this review, we describe the crucial features of cancer stem cells related to tumor relapse and drug resistance, as well as the new therapeutic strategy to target cancer stem cells named "differentiation therapy."
癌症干细胞仅从少量细胞就能生成肿瘤,而分化的癌细胞则不能。癌症干细胞的突出特征是其自我更新以及分化为多种类型癌细胞的能力。癌症干细胞具有几种独特的致瘤能力,包括干细胞信号转导、致瘤性、转移以及对抗癌药物的耐药性,这些能力受基因或表观遗传变化调控。与参与各种发育过程和组织稳态的正常成体干细胞一样,癌症干细胞在癌症起始和进展过程中通过激活多种干细胞信号通路并抑制分化信号通路来维持其自我更新能力。最近,许多研究聚焦于通过调节干细胞信号通路来靶向癌症干细胞以根除恶性肿瘤,其中一些策略的产品正处于临床前和临床试验阶段。在本综述中,我们描述了与肿瘤复发和耐药性相关的癌症干细胞的关键特征,以及靶向癌症干细胞的名为“分化疗法”的新治疗策略。
