To investigate the protective effect and its possible mechanism of ulinastatin on lung injury caused by hip fracture.
Total 24 SD rats were randomly divided into control,hip fracture and hip fracture+ulinastatin groups (=8). The rats of hip fracture and hip fracture+ulinastatin groups made the left hip fracture. The tail vein of rats of hip fracture+ulinastatin group were injected with 50 000 U/kg ustodin before made fracture model. Killed rats 24 h after mold building. Serum mtDNA,tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 in bronchoalveolar lavage fluid,Toll like receptor-9 (TLR9) and nuclear factor (NF)-κB protein expression in lung tissue,wet/dry ratio and pathological scores of lung were detected in all three groups.
Serum mtDNA,TNF-α and IL-6 in bronchoalveolar lavage fluid,TLR9 and NF-κB protein expressions in lung tissue,wet/dry ratio and pathological scores of lung in hip fracture group were significantly higher than those in control group ( <0.05). Ulinastatin significantly reduced the levels of serum mtDNA,TNF-α and IL-6 in bronchoalveolar lavage fluid,TLR9 and NF-κB protein expressions in lung tissue,as well as wet/dry ratio and pathological scores of lung ( <0.05).
Ulinastatin might could provide protection on lung injury in hip fracture,and mtDNA-TLR9-NF-κB pathway may play the role of reducing inflammatory level in the process.
探讨乌司他丁对髋部骨折所致肺损伤的保护作用及其可能机制。
将24只SD大鼠随机分为对照组、髋部骨折组和髋部骨折+乌司他丁组(每组8只)。髋部骨折组和髋部骨折+乌司他丁组大鼠制作左侧髋部骨折模型。髋部骨折+乌司他丁组大鼠在制作骨折模型前经尾静脉注射50 000 U/kg乌司他丁。造模后24 h处死大鼠。检测三组大鼠血清线粒体DNA(mtDNA)、支气管肺泡灌洗液中肿瘤坏死因子-α(TNF-α)和白细胞介素(IL)-6、肺组织中Toll样受体9(TLR9)和核因子(NF)-κB蛋白表达、肺组织湿/干比及病理评分。
髋部骨折组大鼠血清mtDNA、支气管肺泡灌洗液中TNF-α和IL-6、肺组织中TLR9和NF-κB蛋白表达、肺组织湿/干比及病理评分均显著高于对照组(P<0.05)。乌司他丁显著降低了血清mtDNA、支气管肺泡灌洗液中TNF-α和IL-6水平、肺组织中TLR9和NF-κB蛋白表达以及肺组织湿/干比和病理评分(P<0.05)。
乌司他丁可能对髋部骨折所致肺损伤具有保护作用,mtDNA-TLR9-NF-κB通路可能在此过程中发挥降低炎症水平的作用。
