This experiment examined the effect of L. extract on in vitro maturation, in vitro fertilization (IVF) and subsequent developmental competence of mouse oocytes.
Cumulus oocyte complexes (COCs) at germinal vesicle stage were collected from female Naval Medical Research Institute (NMRI) mouse ovaries. The COCs were transferred to maturation medium supplemented with different concentrations of extract. Two trials were carried out to examine the effect of low concentrations (0, 10, 15, 20, 25 μg/ml) and high concentrations (0, 50, 100, 200 μg/ml) of the extract. The maturation rate was recorded. After IVF, embryos were cultured and their developmental process was monitored for 96 h.
Maturation rate and blastocyst formation improved by using low concentrations of the extract; however, no significant increase was observed when compared to the control group. In addition no significant differences were observed in the fertilization rates of oocytes treated with both low and high concentrations compared to the control group. However, among higher concentrations, 100 μg/ml, extract significantly increased both the in vitro maturation rate and in vitro developmental (IVD) competence when compared to the control group ( < 0.05).
It was concluded that natural extracts increase the IVD competence of oocytes. The improved effect on oocyte maturation was dependent on the addition of optimum concentrations of extract to the maturation medium.
本实验研究了L.提取物对小鼠卵母细胞体外成熟、体外受精(IVF)及后续发育能力的影响。
从海军医学研究所(NMRI)雌性小鼠卵巢中收集生发泡期的卵丘卵母细胞复合体(COCs)。将COCs转移至添加不同浓度提取物的成熟培养基中。进行了两项试验,分别检测低浓度(0、10、15、20、25μg/ml)和高浓度(0、50、100、200μg/ml)提取物的作用。记录成熟率。体外受精后,培养胚胎并监测其96小时的发育过程。
使用低浓度提取物可提高成熟率和囊胚形成率;然而,与对照组相比,未观察到显著增加。此外,与对照组相比,用低浓度和高浓度处理的卵母细胞受精率未观察到显著差异。然而,在较高浓度中,与对照组相比,100μg/ml提取物显著提高了体外成熟率和体外发育(IVD)能力(P<0.05)。
得出天然提取物可提高卵母细胞IVD能力的结论。对卵母细胞成熟的改善作用取决于在成熟培养基中添加最佳浓度的提取物。
