To evaluate the potential association of IL-27 polymorphisms with allergic rhinitis(AR) in a Chinese Han population. A case-control study was performed in 445 Chinese AR patients and 691 healthy controls. Three SNPs in the IL-27p28 gene, including rs153109, rs17855750 and rs181206, were detected using a polymerase chain reaction-restriction fragment length polymorphism assay(PCR-RFLP). A significantly increased prevalence of the rs153109 TT genotype and the T allele was found in AR patients, while a decreased prevalence of the CT and CC genotypes and the C allele was found. For rs153109, the TT genotype and the T allele were significantly associated with the risk of AR, but the CT and CC genotypes and the C allele decreased the risk of AR; for rs17855750, the TT genotype and T allele were risk factors for AR, and the GT genotype and G allele provided protection. TTT and TTC haplotypes in the IL-27 p28 gene were positively correlated with AR, while CGT, CTC and CTT haplotypes were associated with a significantly decreased risk of AR. This study indicates that IL-27 p28 polymorphisms rs153109 and rs17855750 are likely involved in AR susceptibility, making them potentially useful genetic biomarkers for AR susceptibility in the Chinese Han population.
为评估白细胞介素-27（IL-27）基因多态性与中国汉族人群变应性鼻炎（AR）的潜在关联。对445例中国AR患者和691例健康对照进行了病例对照研究。采用聚合酶链反应-限制性片段长度多态性分析（PCR-RFLP）检测IL-27 p28基因中的3个单核苷酸多态性（SNP），包括rs153109、rs17855750和rs181206。AR患者中rs153109的TT基因型和T等位基因的患病率显著增加，而CT和CC基因型以及C等位基因的患病率降低。对于rs153109，TT基因型和T等位基因与AR风险显著相关，但CT和CC基因型以及C等位基因降低了AR风险；对于rs17855750，TT基因型和T等位基因是AR的危险因素，而GT基因型和G等位基因具有保护作用。IL-27 p28基因中的TTT和TTC单倍型与AR呈正相关，而CGT、CTC和CTT单倍型与AR风险显著降低相关。本研究表明，IL-27 p28基因多态性rs153109和rs17855750可能与AR易感性有关，使其有可能成为中国汉族人群AR易感性的有用遗传生物标志物。
