Abstract

So-Cheong-Ryong-Tang, which is a standardized Korean medicine of the National Health Insurance, is a traditional prescription for the treatment of allergic rhinitis, bronchitis, and bronchial asthma. Simultaneous analysis and development of SCRT is essential for its stability, efficacy, and risk management. In this study, a simple, reliable, and accurate method using ultrahigh-performance liquid chromatography (UPLC) fingerprinting with a diode array detector (DAD) was developed for the simultaneous analysis. The chromatographic separation of the analytes was performed by an ACQUITY UPLC BEH C18 column (1.7 M, 2.1 × 100 mm, Waters) with a mobile phase of water containing 0.01% (v/v) phosphoric acid and acetonitrile containing 0.01% (v/v) phosphoric acid. The flow rate and detection wavelength were set at 0.4 mL/min and 215, 230, 254, and 280 nm. All calibration curves of the thirteen components showed good linearity ( > 0.999). The limit of detection and limit of quantification ranged 0.001-0.360 and 0.004-1.200 g/mL, respectively. The relative standard deviation (RSD) of intra- and interday was less than 2.60%, and the recoveries were within the range 76.08-103.79% with an RSD value of 0.03-1.50%. The results showed that the developed method was simple, reliable, accurate, sensitive, and precise for the quantification of bioactive components of SCRT.

소청룡용탕은 국민건강보험의 표준화한 한약으로 천식성 비염, 기관지염 및 기관지 천식의 치료를 위한 전통 처방이다. 소청룡용탕의 안정성, 효능 및 위험 관리에 대한 동시 분석 및 개발은 필수적이다. 본 연구에서는 다이오드 어레이 검출기(DAD)를 사용한 초고성능 액체 크로마토그래피(UPLC) 지문을 이용한 간단하고 믿음직하며 정확한 방법을 개발하여 동시 분석을 수행하였다. 분석물의 크로마토그래피 분리를 ACQUITY UPLC BEH C18 컬럼(1.7M, 2.1×100mm, Waters)을 사용하여 0.01%(v/v) 인산이 함유된 물과 0.01%(v/v) 인산이 함유된 아세토니트릴의 유동상을 사용하여 수행하였다. 유속 및 검출 파장은 각각 0.4mL/min 및 215, 230, 254 및 280nm으로 설정되었다. 13가지 성분의 모든 검정 곡선은 양호한 선형성을 보였다(>0.999). 검출 한계 및 정량 한계는 각각 0.001-0.360 및 0.004-1.200μg/mL 범위였다. 일일 내 및 일일 간의 상대 표준 편차(RSD)는 2.60% 미만이며, 회수율은 76.08-103.79% 범위 내에 있으며 RSD 값은 0.03-1.50%였다. 결과는 개발된 방법이 소청룡용탕의 생리활성 성분의 정량에 대해 간단하고 믿음직하며 정확하고 민감하며 정밀하다는 것을 보였다.
