Abstract

A Gram-positive, strictly anaerobic, rod-shaped bacterium, designated YB-7, was isolated from activated sludge of an anaerobic baffled reactor pond in Weizhou terminal wastewater treatment plant, Beihai, Guangxi, China. Strain YB-7 grew at pH 5.0-12.0 (optimum, pH 7.0), 20-45 °C (37 °C) and NaCl concentration of 0-5 % w/v (optimum, 5 %). 16S rRNA gene sequence analysis results showed that strain YB-7 belonged to the genus Clostridium and it was most closely related to Clostridium tetanomorphum DSM 4474 (96.9 % similarity). The DNA-DNA relatedness of strain YB-7 to Clostridium tetanomorphum DSM 4474 was 47.4 %. The DNA G+C content of strain YB-7 was determined to be 32.3 mol%, and the predominant cellar fatty acid (>10 %) was C16 : 0. Polar lipids of strain YB-7 included diphosphatidylglycerol, phosphatidylethylethanolamine, phosphatidylethanolamine, phosphatidylglycerol, phosphatidylcholine, two unidentified aminophospholipids, two unidentified phospholipids and unidentified lipids. The results of this study supported the conclusion that strain YB-7 should be assigned to a new member of the genus Clostridium, for which the name Clostridium beihaiense sp. nov. is proposed. The type strain is YB-7 (=CICC 24109=KCTC 15555).

一株革兰氏阳性、严格厌氧、杆状细菌,被命名为 YB-7,从中国广西北海威州终端污水处理厂厌氧折流池的活性污泥中分离得到。YB-7 菌株在 pH5.0-12.0(最佳 pH7.0)、20-45°C(37°C)和 0-5%(w/v)NaCl 浓度(最佳 5%)下生长。16S rRNA 基因序列分析结果表明,菌株 YB-7 属于梭菌属,与产痉挛梭菌 DSM 4474(相似度 96.9%)最为密切。菌株 YB-7 与产痉挛梭菌 DSM 4474 的 DNA-DNA 同源性为 47.4%。菌株 YB-7 的 DNA G+C 含量为 32.3mol%,主要的细胞脂肪酸(>10%)为 C16:0。YB-7 菌株的极性脂包括双磷脂酰甘油、磷脂酰乙醇胺、磷脂酰乙醇胺、磷脂酰甘油、磷脂酰胆碱、两种未鉴定的氨基磷脂、两种未鉴定的磷脂和未鉴定的脂类。研究结果支持菌株 YB-7 应归属于梭菌属的一个新成员,因此提议将其命名为北海梭菌 sp. nov。其模式菌株为 YB-7(=CICC 24109=KCTC 15555)。
