Failed fertilization after intra-cytoplasmic sperm injection (ICSI) is mainly attributed to failed oocyte activation and can be overcome by artificial oocyte activation (AOA). The present study aims to compare outcomes of ICSI following two different assisted oocyte activation chemical procedures (SrCl and Ionomycin) in sibling oocytes of ICSI candidates.
From March 2015 until February 2016, 105 infertile men with 99-100% abnormal sperm morphology, irrespective of sperm motility, concentration, or origin (semen or testicular) were included in this study. Out of these, 66 couples accepted to be included in the study group (Ionomycin/ SrCl) and 39 couples requested routine AOA procedure (Ionomycin) as external control group. Primary outcomes of this study (fertilization, embryo quality, and post-implantation development) were compared between these groups.
Significantly higher oocyte activation (67.90±3.6% vs. 51.16±3.6%, =0.004) and fertilization (65.23±3.63% vs. 49.65±3.63%, =0.008) rates were observed in sibling oocytes treated with Ionomycin in comparison to the SrCl sibling group. Percentage of top quality embryos was insignificantly higher in SrCl groups compared to the Ionomycin group (29.90±4.27 vs. 20.65±4.05%, =0.26).
Ionomycin may be superior to SrCl for inducing oocyte activation. However, SrCl may be a more efficient means to support the development of better quality embryos following ICSI.
卵胞浆内单精子注射(ICSI)后受精失败主要归因于卵母细胞激活失败,可通过人工卵母细胞激活(AOA)来克服。本研究旨在比较在ICSI候选者的同胞卵母细胞中,两种不同的辅助卵母细胞激活化学方法(SrCl₂和离子霉素)后的ICSI结果。
2015年3月至2016年2月,本研究纳入了105名精子形态异常率为99 - 100%的不育男性,无论其精子活力、浓度或来源(精液或睾丸)如何。其中,66对夫妇同意纳入研究组(离子霉素/SrCl₂),39对夫妇要求采用常规AOA程序(离子霉素)作为外部对照组。比较了这些组之间本研究的主要结果(受精、胚胎质量和植入后发育)。
与SrCl₂同胞组相比,用离子霉素处理的同胞卵母细胞的卵母细胞激活率(67.90±3.6%对51.16±3.6%,P = 0.004)和受精率(65.23±3.63%对49.65±3.63%,P = 0.008)显著更高。与离子霉素组相比,SrCl₂组优质胚胎的百分比略高,但无统计学意义(29.90±4.27对20.65±4.05%,P = 0.26)。
离子霉素在诱导卵母细胞激活方面可能优于SrCl₂。然而,SrCl₂可能是支持ICSI后更好质量胚胎发育的更有效方法。
