Abstract

An innovative analytical method based on high resolution sampling two-dimensional liquid chromatography (HiRes 2D-LC) was established for determination of chlorogenic acid and cynaroside in . A C18 column was used in the first dimension (D)-LC separation with acetonitrile and 0.4% (v/v) phosphoric acid aqueous solution as mobile phases. Five heart cuts of chlorogenic acid and four heart cuts of cynaroside were stored in 2D-LC interface, which was a 5-position-10-port valve equipped with two multiple heart-cutting valves. The stored cuts were sequentially separated in the second dimension (D)-LC. The D separation was carried out on an SB-Phenyl column with acetonitrile and 0.5% (v/v) acetic acid aqueous solution as mobile phases. The results showed that chlorogenic acid peaks in the D were well separated, whereas cynaroside peaks in the D were co-eluted with interferences. The above two targets were accurately quantified through a high resolution sampling mode based on continuous slice cuts of the whole target peaks. The method had good linearity, recovery and repeatability. The HiRes 2D-LC system could be used to improve separation and quantification of (un)targets in traditional Chinese medicine samples.

建立了一种基于高分辨率采样二维液相色谱(HiRes 2D-LC)的创新分析方法,用于测定[具体物质]中的绿原酸和木犀草苷。在一维(1D)-LC分离中使用C18柱,以乙腈和0.4%(v/v)磷酸水溶液作为流动相。绿原酸的5个中心切割馏分和木犀草苷的4个中心切割馏分储存在2D-LC接口中,该接口是一个配备有两个多重中心切割阀的5位10端口阀。储存的馏分在二维(2D)-LC中依次分离。2D分离在SB-苯基柱上进行,以乙腈和0.5%(v/v)乙酸水溶液作为流动相。结果表明,1D中的绿原酸峰分离良好,而2D中的木犀草苷峰与干扰物共洗脱。通过基于整个目标峰连续切片切割的高分辨率采样模式对上述两个目标进行了准确定量。该方法具有良好的线性、回收率和重复性。HiRes 2D-LC系统可用于改善中药样品中(非)目标物的分离和定量。
