Abstract

BACKGROUND/AIM: This study aimed to identify biomarkers for predicting the prognosis of advanced gastric cancer patients who received docetaxel, cisplatin, and S-1 (DCS).

MATERIALS AND METHODS

Gene expression profiles were obtained from the Gene Expression Omnibus database (GSE31811). Gene-Ontology-enrichment and KEGG-pathway analysis were used for evaluating the biological functions of differentially-expressed genes. Protein-protein interaction (PPI) network and Kaplan-Meier survival analyses were employed to assess the prognostic values of hub genes.

RESULTS

A total of 1,486 differentially expressed genes (DEGs) were identified, including 13 up-regulated and 1,473 down-regulated genes. KEGG pathways such as metabolic pathways, cell adhesion molecules (CAMs), PI3K-Akt signaling pathway and pathways in cancer were significantly represented. In the PPI network, the top ten hub genes ranked by degree were GNG7, PLCB1, CALML5, FGFR4, GRB2, JAK3, ADCY7, ADCY9, GNAS and KDR. Five DEGs, including ANTXR1, EFNA5, GAMT, E2F2 and NRCAM, were associated with relapse-free survival and overall survival.

CONCLUSION

ANTXR1, EFNA5, GAMT, E2F2 and NRCAM are potential biomarkers and therapeutic targets for DCS treatment in GC.

从基因表达综合数据库(GSE31811)获取基因表达谱。采用基因本体富集分析和KEGG通路分析评估差异表达基因的生物学功能。利用蛋白质-蛋白质相互作用(PPI)网络和Kaplan-Meier生存分析评估枢纽基因的预后价值。

结果

共鉴定出1486个差异表达基因(DEG),其中13个上调基因和1473个下调基因。显著富集的KEGG通路包括代谢通路、细胞黏附分子(CAM)、PI3K-Akt信号通路和癌症相关通路。在PPI网络中,按度数排名前十的枢纽基因是GNG7、PLCB1、CALML5、FGFR4、GRB2、JAK3、ADCY7、ADCY9、GNAS和KDR。包括ANTXR1、EFNA5、GAMT、E2F2和NRCAM在内的5个DEG与无复发生存率和总生存率相关。

结论

ANTXR1、EFNA5、GAMT、E2F2和NRCAM是GC中DCS治疗的潜在生物标志物和治疗靶点。
