A novel Gram-stain-positive, strictly aerobic, motile, spore-forming and rod-shaped bacterial strain, designated R-3, was isolated from a soil sample obtained from the shore of Lake Panyang, Sichuan Province, PR China. Strain R-3 hydrolysed starch and casein. It could not assimilate d-glucose as a carbon source, or produce acid from d-glucose and l-arabinose. Phylogenetic, phenotypic, chemotaxonomic and molecular studies were performed on the new isolate. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain R-3 was a member of the genus Paenibacillus, exhibiting the highest sequence similarity to Paenibacillus sinopodophylli TEGR-3 (98.4 %). The organism grew at 4-38 °C (optimum, 28-30 °C), at pH 6.0-10.0 (pH 7.0-7.5) and with 0-2.5 % (w/v) NaCl (1 %). The predominant menaquinone was MK-7. Anteiso-C15 : 0 (60.7 %) and C16 : 0 (15.5 %) were the major fatty acids. The cellular polar lipids contained diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, three unidentified aminophospholipids and one unidentified phospholipid. The DNA G+C content of strain R-3 was determined to be 47.0 mol%. The DNA-DNA relatedness between strain R-3 and P. sinopodophylli TEGR-3 was 21.2 %. Based on the results obtained in this study, strain R-3 is considered to represent a novel species of the genus Paenibacillus, for which the name Paenibacillusluteus sp. nov. is proposed. The type strain is R-3 (=CGMCC 1.16135=KCTC 33912).
一株新型革兰氏阳性、严格需氧、能运动、产芽孢的杆状细菌菌株,被命名为 R-3,从中国四川省潘阳湖岸边的土壤样本中分离得到。R-3 菌株可水解淀粉和酪蛋白。它不能同化 d-葡萄糖作为碳源,也不能从 d-葡萄糖和 l-阿拉伯糖产酸。对新分离株进行了系统发育、表型、化学分类和分子研究。基于 16S rRNA 基因序列的系统发育分析表明,R-3 菌株是巨大芽孢杆菌属的一个成员,与巨大芽孢杆菌 TEGR-3(98.4%)的序列相似性最高。该菌在 4-38°C(最佳 28-30°C)、pH 6.0-10.0(pH 7.0-7.5)和 0-2.5%(w/v)NaCl(1%)下生长。主要的menaquinone 是 MK-7。anteiso-C15:0(60.7%)和 C16:0(15.5%)是主要的脂肪酸。细胞极性脂含有双磷脂酰甘油、磷脂酰甘油、磷脂酰乙醇胺、三种未鉴定的氨基磷脂和一种未鉴定的磷脂。R-3 菌株的 DNA G+C 含量为 47.0 mol%。R-3 菌株与巨大芽孢杆菌 TEGR-3 的 DNA-DNA 同源性为 21.2%。基于本研究的结果,R-3 菌株被认为代表了巨大芽孢杆菌属的一个新种,建议将其命名为藤黄微球菌新种。模式菌株为 R-3(=CGMCC 1.16135=KCTC 33912)。
