Abstract

The aim of this study was to isolate a novel esterase from a hypersaline lake by sequence-based metagenomics. The metagenomic DNA was isolated from the enriched hypersaline lake sediment. Degenerate primers targeting the conserved regions of lipolytic enzymes of halophilic microorganisms were used for polymerase chain reaction (PCR) and a whole gene was identified by genome walking. The gene was composed of 783 bp, which corresponds to 260 amino acids with a molecular weight of 28.2 kDa. The deduced amino acid sequence best matched with the esterase from Halomonas gudaonensis with an identity of 91%. Recombinantly expressed enzyme exhibited maximum activity towards pNP-hexanoate with a k value of 12.30 s. The optimum pH and temperature of the enzyme were found as 9 and 30 °C, respectively. The effects of NaCl, solvents, metal ions, detergents and enzyme inhibitors were also studied. In conclusion, a novel enzyme, named as hypersaline lake "Acıgöl" esterase (hAGEst), was identified by sequence-based metagenomics. The high expression level, the ability to maintain activity at cold temperatures and tolerance to DMSO and metal ions are the most outstanding properties of the hAGEst.

本研究旨在通过基于序列的宏基因组学从高盐湖泊中分离一种新型酯酶。从富含有机物的高盐湖泊沉积物中提取宏基因组 DNA。利用针对嗜盐微生物脂酶保守区域的简并引物进行聚合酶链反应(PCR),通过基因组步移法鉴定全长基因。该基因由 783bp 组成,编码 260 个氨基酸,分子量为 28.2kDa。推导的氨基酸序列与盐单胞菌(Halomonas gudaonensis)的酯酶具有最高的同源性,同源性为 91%。重组表达的酶对 pNP-己酸酯表现出最大的活性,k 值为 12.30s。该酶的最适 pH 和温度分别为 9 和 30°C。还研究了 NaCl、溶剂、金属离子、洗涤剂和酶抑制剂的影响。总之,通过基于序列的宏基因组学鉴定了一种新型酶,命名为高盐湖泊“Acıgöl”酯酶(hAGEst)。高表达水平、在低温下保持活性的能力以及对 DMSO 和金属离子的耐受性是 hAGEst 的最显著特性。
