Abstract

OBJECTIVE

The vital role of long noncoding RNAs (lncRNAs) in tumor progression has been identified in numerous studies. In this research, the biological function of lncRNA LINC00052 during the development of cervical cancer was mainly explored.

PATIENTS AND METHODS

LINC00052 expression was detected by quantitative Real-time polymerase chain reaction (qRT-PCR) in cervical cancer tissue samples and cell lines. Moreover, the correlation between LINC00052 expression level and disease-free survival rate of cervical cancer patients was analyzed. In vitro functions of LINC00052 in cervical cancer cells were evaluated by proliferation assay, wound healing assay and transwell assay. In addition, qRT-PCR and Western blot were utilized to explore the underlying mechanism of LINC00052 in mediating the progression of cervical cancer.

RESULTS

LINC00052 expression level was lower in cervical cancer samples than that in adjacent tissues, which was correlated with disease-free survival time. Moreover, cell proliferation, migration and invasion were inhibited through overexpression of LINC00052 in vitro. The mRNA and protein expression of signal transducers and activators of transcription 3 (STAT3) was downregulated after overexpressing LINC00052 in cervical cancer cells. The STAT3 expression level was negatively correlated with the expression of LINC00052 in cervical cancer tissues.

CONCLUSIONS

LINC00052 could repress metastasis and invasion of cervical cancer cell via suppressing STAT3. LINC00052 might be a novel tumor suppressor in cervical cancer.

目的

众多研究已证实长链非编码RNA(lncRNA)在肿瘤进展中发挥着至关重要的作用。本研究主要探讨lncRNA LINC00052在宫颈癌发生发展过程中的生物学功能。

患者与方法

采用定量实时聚合酶链反应(qRT-PCR)检测宫颈癌组织样本及细胞系中LINC00052的表达情况。此外,分析LINC00052表达水平与宫颈癌患者无病生存率之间的相关性。通过增殖实验、伤口愈合实验和Transwell实验评估LINC00052在宫颈癌细胞中的体外功能。另外,利用qRT-PCR和蛋白质免疫印迹法探究LINC00052介导宫颈癌进展的潜在机制。

结果

宫颈癌样本中LINC00052的表达水平低于相邻组织,且与无病生存时间相关。此外,体外过表达LINC00052可抑制细胞增殖、迁移和侵袭。在宫颈癌细胞中过表达LINC00052后,转录信号转导子和激活子3(STAT3)的mRNA和蛋白表达下调。在宫颈癌组织中,STAT3表达水平与LINC00052的表达呈负相关。

结论

LINC00052可通过抑制STAT3来抑制宫颈癌细胞的转移和侵袭。LINC00052可能是宫颈癌中的一种新型肿瘤抑制因子。
