Hesperidin is a well-known flavanone glycoside copiously found in sweet orange and lemon, which was recently reported to possess significant anti-inflammatory, analgesic, antifungal, antiviral, antioxidant, and anticancer activities. Ischemia-reperfusion (I/R) injury is a major problem after renal transplantation. Furthermore, inflammatory responses to I/R exacerbate the resultant renal injury. In the present study, we investigated whether hesperidin exhibits renoprotective effects against I/R-induced acute kidney injury in a rat model.
We fed Sprague-Dawley rats either hesperidin (100 mg/kg/d) or saline. One week later, ischemia was induced by bilateral renal pedicle occlusion for 30 minutes followed by reperfusion. The rats were randomly divided into 3 groups, which were treated as follows: 1. the sham operated group; 2. the I/R group; 3. the I/R-hesperidin group RESULTS: Compared to the sham group, the I/R group had higher expression of blood urea nitrogen and serum creatinine and lower expression of catalase, superoxide dismutase, glutathione peroxidase, antioxidants, and nitric oxide. Compared to the I/R group, the I/R-hesperidin group had higher expression of catalase, superoxide dismutase, glutathione peroxidase, antioxidant, and nitric oxide and lower expression of blood urea nitrogen and serum creatinine.
Hesperidin improved acute renal I/R injury through its antioxidant effects. These findings suggest that hesperidin is a potential therapeutic agent for acute ischemia-induced renal damage.
橙皮苷是一种在甜橙和柠檬中大量存在的著名黄酮糖苷,最近有报道称其具有显著的抗炎、镇痛、抗真菌、抗病毒、抗氧化和抗癌活性。缺血再灌注(I/R)损伤是肾移植后的一个主要问题。此外,对I/R的炎症反应会加剧由此导致的肾损伤。在本研究中,我们调查了橙皮苷在大鼠模型中是否对I/R诱导的急性肾损伤具有肾保护作用。
我们给斯普拉格-道利大鼠喂食橙皮苷(100mg/kg/d)或生理盐水。一周后,通过双侧肾蒂闭塞30分钟诱导缺血,随后进行再灌注。大鼠被随机分为3组,治疗如下:1.假手术组;2.I/R组;3.I/R-橙皮苷组。结果:与假手术组相比,I/R组血尿素氮和血清肌酐表达较高,而过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶、抗氧化剂和一氧化氮表达较低。与I/R组相比,I/R-橙皮苷组过氧化氢酶、超氧化物歧化酶、谷胱甘肽过氧化物酶、抗氧化剂和一氧化氮表达较高,血尿素氮和血清肌酐表达较低。
橙皮苷通过其抗氧化作用改善急性肾I/R损伤。这些发现表明橙皮苷是急性缺血性肾损伤的一种潜在治疗药物。
