Use of the nylon wool column method to separate lymphocytes results in preparations which, while fully viable, have grossly depleted cellular levels of phospholipid, cholesterol and sialic acid. These membrane effects, which relate mainly to T cells, indicate that the method is unsuitable for use in biochemical analysis of lymphocytes.
