Abstract

OBJECTIVE

To investigate the immunomodulatory mechanism by which Yangfei Huoxue decoction (YHD) alleviates bleomycin (BLM)-induced pulmonary fibrosis (PF) in rats.

METHODS

Rats were randomly divided into two time-point groups (day 14 and 28), and each time-point group comprised the following six subgroups: control, BLM, dexamethasone (DXM), YHD high dose (YHD-H), YHD middle dose (YHD-M), and YHD low dose (YHD-L). Haematoxylin and eosin and Masson staining, flow cytometry, enzyme-linked immunosorbent assay, Western blotting and UPLC-QT of analyses were examined.

RESULTS

The results showed that YHD reduced the degree of alveolar inflammation and fibrosis; downregulated the expression of CD28, CD80, CD86, Delta-like 1, Notch2, and Notch3; and upregulated the proportions of Th1/Th2 and Tc1/Tc2. The seven components of YHD were detected.

CONCLUSION

The current study indicates that YHD mainly functions by regulating the immune system and that the molecular mechanism may be related to the regulation of the Notch signaling pathway.

目的

研究杨肺活血汤(YHD)减轻博来霉素(BLM)诱导的大鼠肺纤维化(PF)的免疫调节机制。

方法

将大鼠随机分为两个时间点组(第 14 天和第 28 天),每个时间点组又分为以下六个亚组:对照组、BLM 组、地塞米松(DXM)组、YHD 高剂量(YHD-H)组、YHD 中剂量(YHD-M)组和 YHD 低剂量(YHD-L)组。进行苏木精和伊红染色、马松染色、流式细胞术、酶联免疫吸附试验、Western blot 和 UPLC-QT 分析。

结果

结果表明,YHD 减轻了肺泡炎症和纤维化的程度;下调了 CD28、CD80、CD86、Delta-like 1、Notch2 和 Notch3 的表达;并上调了 Th1/Th2 和 Tc1/Tc2 的比例。检测到 YHD 的七种成分。

结论
