Abstract

OBJECTIVE

Recently, the role of long noncoding RNA (lncRNAs) in tumor progression has attracted much attention. The aim of this study was to investigate the role of lncRNA SNHG16 in the development of Wilms' tumor, and to explore the underlying mechanism.

PATIENTS AND METHODS

Real Time-quantitative Polymerase Chain Reaction (RT-qPCR) was used to detect SNHG16 expression in Wilms' tumor patients' tissues. Function assays, including wound healing assay, and transwell assay, were conducted to detect the changes of biological behaviors in Wilms' tumor cells due to gain or loss of SNHG16. Besides, the luciferase reporter gene assay was performed to explore the underlying mechanism.

RESULTS

The expression level of SNHG16 was significantly up-regulated in Wilms' tumor tissues when compared with adjacent tissues. Cell migration and invasion abilities were significantly repressed via down-regulation of SNHG16. However, opposite results were obtained after up-regulation of SNHG16 in vitro. After the down-regulation of SNHG16, the expression of miR-200a-3p increased significantly. However, the expression of miR-200a-3p was remarkably reduced via up-regulation of SNHG16 in vitro. Furthermore, SNHG16 acted as a competing endogenous RNA via sponging miR-200a-3p in Wilms' tumor.

CONCLUSIONS

SNHG16 induced the metastasis of Wilms' tumor via sponging miR-200a-3p. Our findings might provide a new prospect for the diagnosis and therapy of Wilms' tumor.

目的

近年来,长链非编码RNA(lncRNAs)在肿瘤进展中的作用备受关注。本研究旨在探讨lncRNA SNHG16在肾母细胞瘤发生发展中的作用,并探究其潜在机制。

患者与方法

采用实时定量聚合酶链反应(RT-qPCR)检测肾母细胞瘤患者组织中SNHG16的表达。通过功能实验,包括伤口愈合实验和Transwell实验,检测SNHG16表达上调或下调后肾母细胞瘤细胞生物学行为的变化。此外,进行荧光素酶报告基因实验以探究潜在机制。

结果

与癌旁组织相比,肾母细胞瘤组织中SNHG16的表达水平显著上调。下调SNHG16可显著抑制细胞迁移和侵袭能力。然而,体外上调SNHG16后得到相反结果。下调SNHG16后,miR-200a-3p的表达显著增加。然而,体外上调SNHG16可显著降低miR-200a-3p的表达。此外,在肾母细胞瘤中,SNHG16通过海绵吸附miR-200a-3p发挥竞争性内源RNA的作用。

结论

SNHG16通过海绵吸附miR-200a-3p诱导肾母细胞瘤转移。我们的研究结果可能为肾母细胞瘤的诊断和治疗提供新的前景。
