Abstract

OBJECTIVES

The useful and effective role of exercise program to prevent cardiac tissue apoptosis and fibrosis in ovariectomized type 2 diabetic (T2DM) rats (OVR.D) is well known. The current study aimed to investigate the simultaneous effects of T2DM and swimming plan on the expression of some apoptotic, anti-apoptotic biomarkers and glycogen changes in the cardiac muscle tissue of ovariectomized (OVR) rats.

MATERIALS AND METHODS

Forty rats were randomly sorted into 4 equal categories; sham, OVR, OVR.D and diabetic ovariectomized with an 8 week of swimming plan (OVR.D.E). Lipid profile and miR-133, , Bax, caspase-3 and caspase-8 levels were evaluated in the cardiac tissue.

RESULTS

Ovariectomy significantly (0.05) increased cholesterol, triglyceride, LDL, Bax, caspase-3, caspase-8 and decreased (0.05) HDL, miR-133, in the cardiac tissue and a further reduction in the expression of miR-133, and an enhancement in Bax, caspase-3 and caspase-8 in OVR.D rats was observed (0.01). However, exercise training significantly reversed all the measured parameters (0.05). Also, exercise training improved abnormal tissue structure, fragmentation and irregular form of glycogen granules in the OVR.D.E compared to OVR and OVR.D animals.

CONCLUSION

Exercise training could prevent the cardiac disturbance, enhance the expression of anti-apoptotic markers and decrease apoptotic biomarkers in the hearts of OVR.D animals. Therefore, based on the findings of this study suggested using the exercise's beneficial effects for prevention of the cardiac cell death in OVR.D animals.

目的

运动方案对预防去卵巢2型糖尿病(T2DM)大鼠(OVR.D)心脏组织凋亡和纤维化的有益且有效的作用已广为人知。本研究旨在探讨T2DM和游泳计划对去卵巢(OVR)大鼠心肌组织中一些凋亡、抗凋亡生物标志物表达及糖原变化的联合影响。

材料与方法

40只大鼠随机分为4组,每组数量相等;假手术组、OVR组、OVR.D组和接受为期8周游泳计划的糖尿病去卵巢大鼠组(OVR.D.E组)。评估心脏组织中的血脂谱以及miR - 133、、Bax、半胱天冬酶 - 3和半胱天冬酶 - 8水平。

结果

去卵巢显著(P<0.05)增加了心脏组织中的胆固醇、甘油三酯、低密度脂蛋白、Bax、半胱天冬酶 - 3、半胱天冬酶 - 8,并降低了(P<0.05)高密度脂蛋白、miR - 133、,且观察到OVR.D大鼠中miR - 133、的表达进一步降低,Bax、半胱天冬酶 - 3和半胱天冬酶 - 8增强(P<0.01)。然而,运动训练显著逆转了所有测量参数(P<0.05)。此外,与OVR和OVR.D动物相比,运动训练改善了OVR.D.E组异常的组织结构、糖原颗粒的碎片化和不规则形态。

结论

运动训练可以预防OVR.D动物心脏的紊乱,增强抗凋亡标志物的表达并降低心脏中的凋亡生物标志物。因此,基于本研究结果,建议利用运动的有益作用预防OVR.D动物的心脏细胞死亡。
