To investigate the existence of oxidative stress in the sera of patients with breast cancer and its effects on the consequent breast cancer.
This study included 50 control volunteers, 50 patients with breast cancer, and 50 patients with post-operative breast cancer. Patients with pre-operative cancer were clinically and histopathologically diagnosed for breast carcinoma with stage 0, not having therapeutic history. The control 50 healthy female volunteers had the same socio-economic status, and no history of any cancer. After obtaining consent, venous blood was collected from the volunteers by vein puncture using a 10 mL sterile disposable syringe and needle. About 8 mL of blood was collected, 4 mL of which was poured into a heparinized bulb and 4 mL was allowed to clot. The levels of MDA, NO, GSH, and activities of RBC-SOD (in RBC lysate), NOS, copper and zinc GPx, and CAT, and vitamins A, C, and E metabolites were measured in the sera of each group.
The activities of RBC-SOD and the levels of MDA, NO, as well as the NOS were significantly higher in the sera of all patients with breast cancer as compared with the controls. However, the levels of GSH and vitamins A, C, and E, as well as the activities of copper and zinc GPx and CAT were decreased in patients with breast cancer when compared with the controls.
The study provides further evidence for the presence of oxidative stress in the serum of patients with breast carcinoma. Patients with higher levels of MDA showed deficiencies of antioxidants and trace elements in the serum. A poor dietary antioxidant status and high oxidant levels are associated with the risk of breast cancer, thus suggesting that patients with breast cancer should take nutritive supplements to balance the antioxidant and oxidant levels for better outcomes.
研究乳腺癌患者血清中氧化应激的存在及其对后续乳腺癌的影响。
本研究纳入50名对照志愿者、50名乳腺癌患者和50名乳腺癌术后患者。术前癌症患者经临床和组织病理学诊断为0期乳腺癌,无治疗史。50名健康女性对照志愿者具有相同的社会经济地位,且无任何癌症病史。获得同意后,使用10 mL无菌一次性注射器和针头通过静脉穿刺从志愿者采集静脉血。采集约8 mL血液,其中4 mL注入肝素化采血管,4 mL使其凝固。测定每组血清中丙二醛(MDA)、一氧化氮(NO)、谷胱甘肽(GSH)水平,红细胞超氧化物歧化酶(RBC-SOD,在红细胞裂解物中)、一氧化氮合酶(NOS)、铜锌谷胱甘肽过氧化物酶(GPx)和过氧化氢酶(CAT)活性,以及维生素A、C和E代谢产物水平。

本研究为乳腺癌患者血清中存在氧化应激提供了进一步证据。MDA水平较高的患者血清中抗氧化剂和微量元素缺乏。饮食中抗氧化剂状态不佳和氧化剂水平较高与乳腺癌风险相关,因此提示乳腺癌患者应服用营养补充剂以平衡抗氧化剂和氧化剂水平,从而获得更好的预后。
