The aim of this study was to investigate the effects of administration of a mixture of (MH) on longitudinal bone growth in normal Sprague Dawley (SD) rats. We measured the femur and tibia length, growth plate area, proliferation of chondrocytes, and expression of insulin-like growth factor-1 (IGF-I) and IGF binding protein-3 (IGFBP-3), and Janus kinase 2 (JAK2)/signal transducer and activator of transcription 5 (STAT5) phosphorylation after dietary administration of MH in SD rats for four weeks. The nose-tail length gain and length of femur and tibia increased significantly in the group that received MH for a period of four weeks. We performed H&E staining and Bromodeoxyuridine/5-Bromo-2'-Deoxyuridine (BrdU) staining to examine the effect of dietary administration of MH on the growth plate and the proliferation of chondrocytes and found that MH stimulated the proliferation of chondrocytes and contributed to increased growth plate height during the process of longitudinal bone growth. In addition, serum levels of IGF-1 and IGFBP-3 and expression of IGF-1 and IGFBP-3 mRNAs in the liver and bone were increased, and phosphorylation of JAK2/STAT5 in the liver was increased in the MH groups. Based on these results, we suggest that the effect of MH on longitudinal bone growth is mediated by increased JAK2/STAT5-induced IGF-1 production.
本研究旨在探讨给予混合草药(MH)对正常 SD 大鼠纵向骨生长的影响。我们测量了股骨和胫骨长度、生长板面积、软骨细胞增殖以及胰岛素样生长因子-1(IGF-1)和 IGF 结合蛋白-3(IGFBP-3)的表达,以及接受 MH 饮食治疗的 SD 大鼠 JAK2/STAT5 磷酸化 4 周后的 Janus 激酶 2(JAK2)/信号转导和转录激活因子 5(STAT5)磷酸化。接受 MH 治疗 4 周的大鼠鼻尾长度和股骨、胫骨长度明显增加。我们进行了 H&E 染色和 BrdU 染色,以检查 MH 对生长板和软骨细胞增殖的影响,发现 MH 刺激软骨细胞增殖,有助于在纵向骨生长过程中增加生长板高度。此外,MH 组血清 IGF-1 和 IGFBP-3 水平以及肝脏和骨骼中 IGF-1 和 IGFBP-3 mRNA 的表达增加,肝脏中 JAK2/STAT5 的磷酸化增加。基于这些结果,我们认为 MH 对纵向骨生长的影响是通过增加 JAK2/STAT5 诱导的 IGF-1 产生来介导的。
