Abstract

(1) Background: LncRNAs could be a promising biomarker to predict the prognosis of various cancers. The significance of lncRNA remains unclear in cancer. In this study, we examined the expression level of in the serum of ovarian cancer patients and the functional role of . (2) Methods: Serum samples were obtained from 108 OC patients and 32 normal patients to measure the expression of in the serum. Ovarian cancer cells were used to investigate the role of in cell proliferation, invasion, migration and apoptosis, and the expression of was knocked down using RNA interference. In addition, knockdown cell lines were used in in vivo experiments. (3) Results: The expression of was significantly higher in the serum of OC patients than in that of control patients ( < 0.05). The knockdown of in ovarian cancer cells led to a decrease in cell proliferation, invasion and migration and an increase in apoptosis. knockdown also reduced the expression of epithelium-mesenchymal metastasis (EMT) genes. (4) Conclusion: These findings highlight the clinical significance of in predicting the prognosis of OC patients and suggest its potential in promoting tumour aggressiveness by the regulation of EMT-related mechanisms.

(1) 背景:长链非编码RNA(lncRNAs)可能是预测各种癌症预后的一种有前景的生物标志物。lncRNA在癌症中的意义仍不清楚。在本研究中,我们检测了卵巢癌患者血清中lncRNA的表达水平及其功能作用。(2) 方法:从108例卵巢癌患者和32例正常患者中获取血清样本,以检测血清中lncRNA的表达。利用卵巢癌细胞研究lncRNA在细胞增殖、侵袭、迁移和凋亡中的作用,并使用RNA干扰技术敲低lncRNA的表达。此外,将lncRNA敲低的细胞系用于体内实验。(3) 结果:卵巢癌患者血清中lncRNA的表达明显高于对照患者(P<0.05)。卵巢癌细胞中lncRNA的敲低导致细胞增殖、侵袭和迁移减少,凋亡增加。lncRNA敲低还降低了上皮-间质转化(EMT)基因的表达。(4) 结论:这些发现突出了lncRNA在预测卵巢癌患者预后中的临床意义,并表明其通过调节EMT相关机制促进肿瘤侵袭性的潜力。
