Abstract

OBJECTIVE

To observe the effect of electroacupuncture(EA) on proangiogenesis process and protein turn-over in a mouse model of sarcopenia, so as to explore its potential molecular mechanism anti-aging.

METHODS

Fourteen 30-week-old male SAMP8 mice were randomly divided into a model group (=7) and an EA group (=7). Seven anti-rapidly aging SAMR1 mice of the same age were used as the control group (=7). EA (1 mA, 4 Hz) was applied to bilateral "Zusanli"(ST36) and "Yanglingquan"(GB34) for 20 minutes each time once a day, 6 times a week for 4 weeks. The exhausted running platform was used to test the sports function. Gastrocnemius muscle mass and relative ratio of gastrocnemius muscle mass to body mass were measured. HE staining and transmission electron microscope were used to observe the morphology, and the cross-sectional area of gastrocnemius muscle was calculated. Relative protein expressions of protein kinase B (AKT) , phosphorylated (p) -AKT, mammalian target of rapamycin (mTOR) , p-mTOR, p70 ribosomal protein S6 kinase (p70S6K) , p-p70S6K,hypoxia inducible factor-1α (HIF-1α) and relative mRNA expressions of HIF-1α, vascular endothelial growth factor A (VEGF-A) , muscle RING finger-1 (MuRF-1) and muscle atrophy F-box (MAFbx) were detected by Western blot and real-time fluorescence quantitative PCR, seperatively.

RESULTS

Compared with the control group, the running time and distance, body mass and gastrocnemius mass, and the ratio of gastrocnemius mass to body mass decreased(<0.01, <0.05), cross-sectional area of gastrocnemius, related protein expression of p-AKT,p-mTOR, p-p70S6K and HIF-1α, mRNA expression of HIF-1α and VEGF-A decreased (<0.01), while mRNA expression of MuRF1 and MAFbx increased (<0.01) in the model group. Following EA intervention, the running time and distance, body mass and gastrocnemius mass and the ratio of gastrocnemius mass to body mass increased (<0.05), cross-sectional area of gastrocnemius, related protein expression of p-AKT,p-mTOR, p-p70S6K and HIF-1α, mRNA expression of HIF-1α and VEGF-A were significantly up-regulated (<0.01), mRNA expression of MuRF1 and MAFbx down-regulated (<0.01, <0.05) in the EA group compared with the model group.

CONCLUSION

EA may delay the aging muscle atrophy in mice by regulating the gastrocnemius muscle's proangiogenesis process and protein turnover.

目的

观察电针(EA)对少肌症小鼠模型血管生成过程和蛋白质周转的影响,以探讨其抗衰老的潜在分子机制。

方法

将14只30周龄雄性SAMP8小鼠随机分为模型组(n = 7)和电针组(n = 7)。选取7只同龄抗快速衰老的SAMR1小鼠作为对照组(n = 7)。电针(1 mA,4 Hz)刺激双侧“足三里”(ST36)和“阳陵泉”(GB34),每次20分钟,每天1次,每周6次,共4周。采用疲劳跑步平台测试运动功能。测量腓肠肌质量及腓肠肌质量与体重的相对比值。采用苏木精-伊红(HE)染色和透射电子显微镜观察形态,并计算腓肠肌横截面积。分别采用蛋白质免疫印迹法和实时荧光定量PCR检测蛋白激酶B(AKT)、磷酸化(p)-AKT、雷帕霉素靶蛋白(mTOR)、p-mTOR、p70核糖体蛋白S6激酶(p70S6K)、p-p70S6K、缺氧诱导因子-1α(HIF-1α)的相对蛋白表达以及HIF-1α、血管内皮生长因子A(VEGF-A)、肌肉环状指蛋白-1(MuRF-1)和肌肉萎缩F盒蛋白(MAFbx)的相对mRNA表达。

结果

与对照组相比,模型组小鼠的跑步时间和距离、体重和腓肠肌质量以及腓肠肌质量与体重的比值降低(P<0.01,P<0.05),腓肠肌横截面积、p-AKT、p-mTOR、p-p70S6K和HIF-1α的相关蛋白表达、HIF-1α和VEGF-A的mRNA表达降低(P<0.01),而MuRF1和MAFbx的mRNA表达升高(P<0.01)。电针干预后,与模型组相比,电针组小鼠的跑步时间和距离、体重和腓肠肌质量以及腓肠肌质量与体重的比值增加(P < 0.05),腓肠肌横截面积、p-AKT、p-mTOR、p-p70S6K和HIF-1α的相关蛋白表达、HIF-1α和VEGF-A的mRNA表达显著上调(P<0.01),MuRF1和MAFbx的mRNA表达下调(P<0.01,P<0.05)。

结论

电针可能通过调节腓肠肌的血管生成过程和蛋白质周转来延缓小鼠衰老性肌肉萎缩。
