Abstract

Osteoarthritis (OA), one of the most common motor system disorders, is a degenerative disease involving progressive joint destruction caused by a variety of factors. At present, OA has become the fourth most common cause of disability in the world. However, the pathogenesis of OA is complex and has not yet been clarified. Long non-coding RNA (lncRNA) refers to a group of RNAs more than 200 nucleotides in length with limited protein-coding potential, which have a wide range of biological functions including regulating transcriptional patterns and protein activity, as well as binding to form endogenous small interference RNAs (siRNAs) and natural microRNA (miRNA) molecular sponges. In recent years, a large number of lncRNAs have been found to be differentially expressed in a variety of pathological processes of OA, including extracellular matrix (ECM) degradation, synovial inflammation, chondrocyte apoptosis, and angiogenesis. Obviously, lncRNAs play important roles in regulating gene expression, maintaining the phenotype of cartilage and synovial cells, and the stability of the intra-articular environment. This article reviews the results of the latest research into the role of lncRNAs in a variety of pathological processes of OA, in order to provide a new direction for the study of OA pathogenesis and a new target for prevention and treatment. 骨关节炎(OA)是最常见的运动系统疾病之一,是一种由多种因素导致关节进行性破坏的退行性疾病。目前,OA已成为全球第四大致残原因。然而,OA的发病机制复杂,尚未完全阐明。长链非编码RNA(lncRNA)是指一组长度超过200个核苷酸、蛋白质编码潜能有限的RNA,其具有广泛的生物学功能,包括调节转录模式和蛋白质活性,以及结合形成内源性小干扰RNA(siRNA)和天然微小RNA(miRNA)分子海绵。近年来,大量lncRNA被发现在OA的多种病理过程中差异表达,包括细胞外基质(ECM)降解、滑膜炎症、软骨细胞凋亡和血管生成。显然,lncRNA在调节基因表达、维持软骨和滑膜细胞表型以及关节内环境稳定性方面发挥着重要作用。本文综述了lncRNA在OA多种病理过程中作用的最新研究结果,以期为OA发病机制的研究提供新方向,为预防和治疗提供新靶点。引用本文:2021;10(2):122-133。
