of Rutaceae, a Taiwan folk medicine, is used as an analgesic and anti-inflammatory herb. Cyclohexylamine (CHA) is an active compound from Previous reports indicate CHA contracts rat vas deferens. However, the contractile mechanism of CHA on rat vas deferens was not yet reported. The purpose of this study was to investigate the contractile mechanism of CHA on rat epididymal portion of vas deferens.
Male S.D. rats weighting between 200 g to 250 g were used. The epididymal portion of vas deferens was isolated and was added with various concentrations of serotonin, serotonin antagonists and CHA.
Serotonin elicited dose-dependent (1 × 10M1 × 10M) contractions on rat epididymal vas deferens, which were inhibited by pretreatment with ketanserin (1 × 10 M ~ 1 × 10 M), methysergide (1 × 10 M) and propranolol (1 × 10 M), respectively. CHA elicited dose-dependent (1 × 10M1 × 10M) contractions on rat epididymal vas deferens. The contractions of CHA (1 × 10M) on epididymal vas deferens were enhanced by serotonin in a dose-dependent manner. Methysergide (1 × 10 ~1 × 10 M) did not affect the contractions evoked by CHA. However, the CHA elicited contraction was almost completely inhibited by ketanserin (1 × 10 M) and was enhanced by propranolol. The effect of propranolol at the concentration of 1 × 10 M on CHA was likely as CHA at high concentration alone.
From the above results, the contraction evoked by CHA on the isolated rat epididymal vas deferens might be mediated by serotonergic receptors through 5-HT subtype.
芸香科植物,一种台湾民间药物,用作止痛和抗炎草药。环己胺(CHA)是该植物的一种活性化合物。先前的报告表明CHA可使大鼠输精管收缩。然而,CHA对大鼠输精管的收缩机制尚未见报道。本研究的目的是探讨CHA对大鼠附睾段输精管的收缩机制。
使用体重在200克至250克之间的雄性SD大鼠。分离出附睾段输精管,并添加不同浓度的5-羟色胺、5-羟色胺拮抗剂和CHA。
5-羟色胺对大鼠附睾输精管产生剂量依赖性(1×10⁻⁹M1×10⁻⁵M)收缩,分别被酮色林(1×10⁻⁹M1×10⁻⁵M)、麦角新碱(1×10⁻⁶M)和普萘洛尔(1×10⁻⁶M)预处理所抑制。CHA对大鼠附睾输精管产生剂量依赖性(1×10⁻⁹M1×10⁻⁵M)收缩。5-羟色胺以剂量依赖性方式增强CHA(1×10⁻⁶M)对附睾输精管的收缩作用。麦角新碱(1×10⁻⁶1×10⁻⁵M)不影响CHA引起的收缩。然而,CHA引起的收缩几乎完全被酮色林(1×10⁻⁵M)抑制,并被普萘洛尔增强。普萘洛尔在1×10⁻⁶M浓度下对CHA的作用可能与单独高浓度的CHA相同。
从上述结果来看,CHA对离体大鼠附睾输精管引起的收缩可能是通过5-羟色胺能受体经5-HT亚型介导的。
