Urine cell-free DNA (cfDNA) is a new type of liquid biopsy biomarker used in tumours and allograft injury detection but is highly susceptible to degradation by the high nuclease activity of urine. This study presents a newly developed urine cfDNA preservation solution (AlloU), efficient for examining allograft injury in kidney transplant recipients (KTx).
We established urine-preserve solution called AlloU based on the response-surface methodology, with two commercial collection reagents (Streck and K2 EDTA preservation solution) included for analysis. A total of 120 urine samples from KTx patients, including morning, nocturnal and random urine from specific storage time were subjected to investigation. The urine total cfDNA concentration was quantified by fluorometry, fragment distribution was analysed by qPCR, and donor-derived cfDNA (ddcfDNA) was detected by next-generation sequencing.
Urine total cfDNA concentration and fragment size of samples preserved with AlloU and Streck did not change significantly within 5 days whereas the ddcfDNA also did not change significantly within 7 days. However, compared with EDTA, the total cfDNA concentration increased significantly on the third day. When compare with different urine types, it was found that samples preserved with AlloU showed no significant differences in total cfDNA concentration, fragment size, and ddcfDNA concentration, however, the SD for morning urine was significantly smaller in total cfDNA and ddcfDNA concentration.
To the best of our knowledge, this is the first report to verify the dynamics of urine cfDNA in KTx, especially in the analysis impact of different urine types on cfDNA detection.
尿液无细胞 DNA(cfDNA)是一种新型的液体活检生物标志物,用于检测肿瘤和移植物损伤,但易受到尿液中高核酸酶活性的降解。本研究提出了一种新开发的尿液 cfDNA 保存液(AlloU),可有效检测肾移植受者(KTx)的移植物损伤。
我们基于响应面法建立了一种名为 AlloU 的尿液保存液,并纳入了两种商业采集试剂(Streck 和 K2 EDTA 保存液)进行分析。共收集了 120 例 KTx 患者的尿液样本,包括特定储存时间的晨尿、夜尿和随机尿。采用荧光法定量检测尿液总 cfDNA 浓度,qPCR 分析片段分布,下一代测序检测供体来源 cfDNA(ddcfDNA)。
AlloU 和 Streck 保存的尿液样本中的总 cfDNA 浓度和片段大小在 5 天内无明显变化,ddcfDNA 也在 7 天内无明显变化。然而,与 EDTA 相比,cfDNA 总浓度在第 3 天显著增加。与不同尿液类型相比,发现 AlloU 保存的样本在总 cfDNA 浓度、片段大小和 ddcfDNA 浓度方面无显著差异,但晨尿的总 cfDNA 和 ddcfDNA 浓度的 SD 明显较小。
