Abstract

Regulation of extracellular Ca influx by neuronal activity is a key mechanism underlying synaptic plasticity. At the neuronal synapse, activity-dependent Ca entry involves N-methyl-D-aspartate receptors (NMDARs) and voltage-gated calcium channels (VGCCs); the relationship between NMDARs and VGCCs, however, is poorly understood. Here, we report that neuronal activity rapidly (1h) regulates recruitment of R-type VGCCs in hippocampal neurons through synaptic NMDAR signalling. This finding reveals a link between two key neuronal signalling pathways, suggesting a feedback mode for regulation of synaptic Ca signalling.

神经元活动对细胞外钙内流的调节是突触可塑性的关键机制。在神经元突触处,依赖活动的钙内流涉及N-甲基-D-天冬氨酸受体(NMDARs)和电压门控钙通道(VGCCs);然而,NMDARs与VGCCs之间的关系却知之甚少。在此,我们报告神经元活动通过突触NMDAR信号通路快速(1小时)调节海马神经元中R型VGCCs的募集。这一发现揭示了两条关键神经元信号通路之间的联系,提示了一种调节突触钙信号的反馈模式。
