The objective of the current study was to evaluate the effect of autologous platelet-rich plasma (PRP) addition into soybean lecithin based extender on buck semen at post-thaw. Semen samples were collected from eight Saanen buck, and each semen sample was split into four equal aliquots and diluted with different concentrations of PRP supplemented extenders [no PRP (control), 0.5 × 10/ml PRP, 1 × 10/ml PRP, or 2 × 10/ml PRP]. Motility, plasma membrane functional integrity, acrosome integrity, mitochondrial membrane potential, DNA integrity and malondialdehyde concentrations (MDA) were measured and analyzed at post-thaw. The results showed that 2 × 10/ml PRP group had a positive effect on motility (62.41 ± 4.24), membrane functional integrity (71.11 ± 2.90), mitochondrial membrane potential (69.70 ± 1.99), DNA integrity (7.22 ± 0.93) and MDA levels (2.56 ± 0.73) at post-thaw (P < 0.05). The results of the study demonstrated that autologous PRP has a protective effect on cryopreservation of buck spermatozoa and the fertility effects are worthy of further study.
本研究的目的是评估在解冻后将自体富血小板血浆(PRP)添加到大豆卵磷脂基础稀释液中对雄鹿精液的影响。从 8 只 Saanen 雄鹿中收集精液样本,每个精液样本均分为 4 等份,并分别用不同浓度添加 PRP 的稀释液稀释[无 PRP(对照)、0.5×10/ml PRP、1×10/ml PRP 或 2×10/ml PRP]。在解冻后对活力、质膜功能完整性、顶体完整性、线粒体膜电位、DNA 完整性和丙二醛浓度(MDA)进行测量和分析。结果表明,2×10/ml PRP 组对活力(62.41±4.24)、膜功能完整性(71.11±2.90)、线粒体膜电位(69.70±1.99)、DNA 完整性(7.22±0.93)和 MDA 水平(2.56±0.73)解冻后有积极影响(P<0.05)。研究结果表明,自体 PRP 对雄鹿精子冷冻保存具有保护作用,其生育效果值得进一步研究。
