As traditional Chinese medicine (TCM), nyctinastic herbs have been used in treating insomnia in China since ancient times according to its similar circadian rhythm as human beings. However, the pharmacodynamic characteristics and mechanism of these herbs have not been explored in depth.
In the study, we chose He Huan Pi ( Durazz.), Ye Jiao Teng ( Thunb.), Bai He ( Baker), and Lianzi ( Gaertn) to form a TCM compound decoction [nyctinastic herb decoction (NHD)] and to investigate its sedative and hypnotic effect on para-chlorophenylalanine (PCPA)-induced insomnia rodents by pentobarbital-induced sleep test (PIST), behavior test [including locomotor activity (LMA), forced swim test (FST), tail suspension test (TST)] and electroencephalograph (EEG). The expression of neurotransmitters were detected to explain the possible mechanism of NHD.
NHD was found to have good sedative effects on reducing the moving distance, prolonging sleep time, improving the sleep quality and depression status. NHD attenuated the insomniac effect of PCPA by increasing the level of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA), and reducing the level of dopamine (DA), norepinephrine (NE), acetylcholine (ACh) in the hypothalamus.
The findings of the present study confirmed the sedative and hypnotic effect of NHD, and clarified its possible mechanism from neurotransmitters.
作为传统中药,夜交藤类草药因其与人类相似的昼夜节律,自古以来就在中国用于治疗失眠。然而,这些草药的药效学特征和作用机制尚未得到深入研究。
在本研究中,我们选用合欢皮(豆科)、夜交藤(蓼科)、百合(百合科)和莲子(睡莲科)组成一种中药复方汤剂[夜交藤类草药汤剂(NHD)],并通过戊巴比妥诱导睡眠试验(PIST)、行为学试验[包括自发活动(LMA)、强迫游泳试验(FST)、悬尾试验(TST)]和脑电图(EEG),研究其对对氯苯丙氨酸(PCPA)诱导的失眠啮齿动物的镇静催眠作用。检测神经递质的表达以解释NHD可能的作用机制。
发现NHD具有良好的镇静作用,可减少活动距离、延长睡眠时间、改善睡眠质量和抑郁状态。NHD通过增加下丘脑5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)水平,降低多巴胺(DA)、去甲肾上腺素(NE)、乙酰胆碱(ACh)水平,减轻PCPA的失眠作用。
本研究结果证实了NHD的镇静催眠作用,并从神经递质方面阐明了其可能的作用机制。
