Abstract

Duck short beak and dwarfism syndrome (SBDS) is a viral infectious disease caused by novel goose parvovirus (NGPV), which has been responsible for serious economic losses to the Chinese duck industry in recent years. Currently, there is no effective vaccine against this disease. In this study, we developed an inactivated virus vaccine candidate for SBDS based on NGPV strain DS15 isolated from a duck in China. Immune efficacy was evaluated in 112 ducks, which were randomly divided into vaccination, challenge-control, vaccination-challenge, and blank control groups (28 per group). Clinical characteristics, antibodies, virus excretion, viremia, and pathological changes were monitored. No morbidity or death was observed in the immunized ducks, which showed normal weight and a good mental state. High levels of serum antibodies (optical density at 450 nm of ~ 0.63) were detected in ducks immunized with the inactivated vaccine at 7 days post-vaccination (dpv), and the titer of virus-neutralizing antibodies increased from 1:2 to 1:2 from 7 to 42 dpv. Measurement of the viral load in anal swab, serum, and tissue samples showed that vaccination significantly inhibited the replication of NGPV in immunized ducks. Moreover, NGPV could not be isolated from the spleens of immunized or vaccinated and challenged ducks. Collectively, these results demonstrate that the newly developed inactivated NGPV vaccine, administered in an oil emulsion adjuvant, possesses good immunogenicity and represents a potentially powerful tool for SBDS prevention and control.

鸭短喙侏儒综合征(SBDS)是一种由新型鹅细小病毒(NGPV)引起的病毒性传染病,近年来已对中国养鸭业造成严重的经济损失。目前,尚无针对该病的有效疫苗。本研究以中国鸭源 NGPV 分离株 DS15 为候选疫苗株,研制了一种鸭短喙侏儒综合征灭活疫苗。将 112 只鸭随机分为免疫组、攻毒对照组、免疫攻毒组和空白对照组(每组 28 只),评估了该疫苗的免疫效果。监测了临床特征、抗体、病毒排出、病毒血症和组织病理学变化。免疫鸭未出现发病率或死亡率,体重正常,精神状态良好。免疫鸭在免疫后 7 天(dpv)血清抗体水平(450nm 处的光密度值约为 0.63)较高,病毒中和抗体滴度从 7dpv 的 1:2 升至 42dpv 的 1:2。分析肛拭子、血清和组织样本中的病毒载量表明,疫苗接种显著抑制了免疫鸭体内 NGPV 的复制。此外,免疫或免疫攻毒鸭的脾脏中均未分离到 NGPV。综上结果表明,新型 NGPV 油乳剂灭活疫苗具有良好的免疫原性,为防控鸭短喙侏儒综合征提供了一种有潜力的工具。
