Abstract

INTRODUCTION

Liver carcinoma is one of the most common cancers in the world and remains one of the most difficult cancers to treat. Hepatocellular cancer is the most important type of liver cancer (90%). RING Finger 44 (RNF44) is one of the E3 ligases, which play an important role in substrate recognition. It was also reported that RING Finger 44 was connected with resistant melanoma. But the relationship between RNF44 and HCC remained unknown.

MATERIALS AND METHODS

To analyze the role of RING Finger 44 gene in hepatocellular carcinoma, we used bioinformatics to analyze the expression level, genetic changes, immunohistochemistry, immune infiltration, diagnostic value, survival, and functional enrichment of RING Finger 44.

RESULTS

Through analyzing The Genotype-Tissue Expression and The Cancer Genome Atlas databases, we found that the expression level of RING Finger 44 was significantly increased in hepatocellular carcinoma tissues. Meanwhile, the expression of RING Finger 44 was connected with immune cell infiltration and survival time, and the expression level of RING Finger 44 could perform as a useful diagnostic and prognostic index. The functional enrichment analysis of RING Finger 44 provided some possible pathways of RING Finger 44 in hepatocellular carcinoma, which provided an important direction for the further experiments in vitro or in vivo.

CONCLUSIONS

RING Finger 44, the high expression level of which predicts poor prognosis, is a potential oncogene in hepatocellular carcinoma.

肝癌是世界上最常见的癌症之一,也是最难治疗的癌症之一。肝细胞癌是最重要的肝癌类型(90%)。RING 指蛋白 44(RNF44)是一种 E3 连接酶,在底物识别中发挥重要作用。有报道称 RING 指蛋白 44 与耐药性黑色素瘤有关。但 RNF44 与 HCC 之间的关系尚不清楚。

材料与方法

为了分析 RING 指蛋白 44 基因在肝细胞癌中的作用,我们使用生物信息学方法分析了 RING 指蛋白 44 的表达水平、遗传变化、免疫组化、免疫浸润、诊断价值、生存和功能富集。

结果

通过分析基因型-组织表达和癌症基因组图谱数据库,我们发现 RING 指蛋白 44 在肝癌组织中的表达水平显著升高。同时,RING 指蛋白 44 的表达与免疫细胞浸润和生存时间有关,RING 指蛋白 44 的表达水平可以作为有用的诊断和预后指标。RING 指蛋白 44 的功能富集分析为 RING 指蛋白 44 在肝细胞癌中的可能途径提供了一些线索,为进一步的体外或体内实验提供了重要方向。

结论

RING 指蛋白 44 的高表达水平预示着预后不良,是肝细胞癌的潜在癌基因。
