To reveal the expression level of cystathionine β-synthase (CBS) in adenocarcinoma of esophagogastric junction (AEG) and discuss the relationship between CBS expression level and tumor microvascular density (MVD), clinical features and prognosis.
Paraffin samples from 214 patients with AEG were selected to make pathological microchips. Immunohistochemistry was performed based on the microchips to detect the expression level of CBS and microvascular density (MVD) in cancer tissues and adjacent control tissues. Relationships between expression level of CBS and MVD, clinical characteristics and prognosis were analyzed.
In total, 214 AEG cases were classified into three groups: CBS negative staining (=26), low staining (=44), and high staining (=144). Quantitative alterations in CBS and CD31 expression were explored using immunohistochemistry. The 5-year recurrence rate of enrolled patients was followed up and found that CBS expression was significantly increased in tumor tissue compared with adjacent non-tumor tissue (<0.0001). There were significant differences in microvascular density between the groups with negative and high CBS staining (<0.0001), and between the groups with low and high CBS staining (<0.0001). Univariate analysis revealed significant differences in tumor stage (<0.0001), T stage (=0.008), N stage (=0.028), differentiation degree (=0.037), and 5-year survival (=0.0034) among the three groups. Multivariate logic regression analysis showed that increased CBS scores were associated with an increased probability of 5-year recurrence (=0.018). Finally, different CBS expression levels were associated with disease-free survival in AEG patients.
CBS expression level is closely related to microvascular density and tumor stage in AEG. High level of CBS not only accelerates tumor angiogenesis but also affects patient's survival and prognosis.

选取214例AEG患者的石蜡样本制作病理微芯片。基于微芯片进行免疫组织化学检测癌组织及癌旁对照组织中CBS的表达水平和微血管密度(MVD)。分析CBS表达水平与MVD、临床特征及预后的关系。
214例AEG病例共分为三组:CBS阴性染色(=26例)、低染色(=44例)和高染色(=144例)。采用免疫组织化学方法探讨CBS和CD31表达的定量变化。对纳入患者进行5年复发率随访,发现肿瘤组织中CBS表达较癌旁非肿瘤组织显著升高(<0.0001)。CBS阴性染色组与高染色组之间微血管密度存在显著差异(<0.0001),CBS低染色组与高染色组之间也存在显著差异(<0.0001)。单因素分析显示三组间肿瘤分期(<0.0001)、T分期(=0.008)、N分期(=0.028)、分化程度(=0.037)和5年生存率(=0.0034)存在显著差异。多因素逻辑回归分析显示CBS评分升高与5年复发概率增加相关(=0.018)。最后,不同CBS表达水平与AEG患者的无病生存期相关。
CBS表达水平与AEG的微血管密度和肿瘤分期密切相关。高水平的CBS不仅加速肿瘤血管生成,还影响患者的生存和预后。
