Abstract

Hydrogen peroxide (HO) is a main member of reactive oxygen species (ROS) in cells that has a significant impact on various physiological and pathological processes of organisms. Here, we designed and synthesized a new type of fluorescent probe Rhodol-OAc for the specific detection of HO. The probe had good water solubility, high selectivity and sensitivity to HO, low cytotoxicity, excellent mitochondrial targeting ability, It was successfully applied in the imaging of exogenous and endogenous HO in living cells. In addition, theoretical calculations were carried out to clarify the luminescence mechanism of the probe. More importantly, we successfully applied the probe to indirectly detect xanthine and glucose, the metabolism of which generates HO, and achieved satisfactory results.

过氧化氢(HO)是细胞内活性氧(ROS)的主要成员,对生物体的各种生理和病理过程都有重要影响。在这里,我们设计并合成了一种新型的荧光探针 Rhodol-OAc,用于特异性检测 HO。该探针具有良好的水溶性、对 HO 具有高选择性和灵敏度、低细胞毒性、优异的线粒体靶向能力,成功应用于活细胞中外源和内源性 HO 的成像。此外,还进行了理论计算以阐明探针的发光机制。更重要的是,我们成功地应用该探针间接检测黄嘌呤和葡萄糖的代谢产物 HO,取得了令人满意的结果。
