This study aimed to reveal the classical signal pathways and important potential targets of traditional Chinese medicine (TCM) for treating Alzheimer's disease (AD), and provide support for further investigation on TCM and its active ingredients.
Literature survey was conducted using PubMed, Web of Science, Google Scholar, CNKI, and other databases, with "Alzheimer's disease," "traditional Chinese medicine," "medicinal herb," "Chinese herb," and "natural plant" as the primary keywords.
TCM could modulate signal pathways related to AD pathological progression, including NF-κB, Nrf2, JAK/STAT, ubiquitin-proteasome pathway, autophagy-lysosome pathway-related AMPK/mTOR, GSK-3/mTOR, and PI3K/Akt/mTOR, as well as SIRT1 and PPARα pathway. It could regulate crosstalk between pathways through a multitarget, thus maintaining chronic inflammatory interaction balance, inhibiting oxidative stress damage, regulating ubiquitin-proteasome system function, modulating autophagy, and eventually improving cognitive impairment in patients with AD.
TCM could be multilevel, multitargeted, and multifaceted to prevent and treat AD. In-depth research on the prevention and treatment of AD with TCM could provide new ideas for exploring the pathogenesis of AD and developing new anti-AD drugs.
本研究旨在揭示中医药治疗阿尔茨海默病(AD)的经典信号通路和重要潜在靶点,为进一步研究中医药及其活性成分提供依据。
通过 PubMed、Web of Science、Google Scholar、CNKI 等数据库进行文献检索,以“阿尔茨海默病”“中医药”“药用植物”“中药”“天然植物”等为主要关键词。
中医药可调节与 AD 病理进展相关的信号通路,包括 NF-κB、Nrf2、JAK/STAT、泛素-蛋白酶体通路、自噬-溶酶体通路相关的 AMPK/mTOR、GSK-3/mTOR 和 PI3K/Akt/mTOR,以及 SIRT1 和 PPARα通路。它可以通过多靶点调节通路之间的相互作用,从而维持慢性炎症相互作用的平衡,抑制氧化应激损伤,调节泛素-蛋白酶体系统功能,调节自噬,最终改善 AD 患者的认知障碍。
中医药可以多层次、多靶点、多方位防治 AD。深入研究中医药防治 AD 可以为探索 AD 的发病机制和开发新的抗 AD 药物提供新的思路。
