Abstract

OBJECTIVE

We investigated the relationships between interleukin- (IL-) 1 and IL-1 receptor antagonist (IL-1Ra) gene polymorphism and plasma levels in patients with diabetic nephropathy (DN).

METHODS

The genotype and allele frequency distribution of IL-1 and IL-1Ra in 61 patients with DN and 48 healthy controls (HCs) were determined by kompetitive allele-specific PCR (KASP), and the plasma concentrations of IL-1 and IL-1Ra in DN patients and HCs were measured by enzyme-linked immunosorbent assays (ELISA).

RESULTS

Significant differences were detected in the distribution of IL-1 (-511C/T) genotype and allele frequencies between the DN and HC groups ( < 0.05), with the T genotype being more frequent in DN patients than HCs (OR = 2.84, 95% CI: 1.489-5.416). The IL-1 (+3953C/T) and IL-1Ra (+8006C/T) genotypes and allele frequencies were not significantly different between the two groups ( > 0.05). The plasma IL-1 level was significantly higher ( < 0.01), while the plasma IL-1Ra concentration was significantly lower in the DN group than the HC group ( < 0.05). Furthermore, the plasma IL-1 level was significantly different between IL-1 (-511C/T) locus variants ( < 0.05).

CONCLUSION

The IL-1 (-511C/T) gene polymorphism was significantly associated with DN risk in the population of northern Guangxi, China, and the T allele maybe responsible for genetic susceptibility to DN.

目的

研究白细胞介素-1(IL-1)和白细胞介素-1 受体拮抗剂(IL-1Ra)基因多态性与糖尿病肾病(DN)患者血浆水平的关系。

方法

采用 kompetitive allele-specific PCR(KASP)法检测 61 例 DN 患者和 48 例健康对照(HC)的 IL-1 和 IL-1Ra 基因型和等位基因频率分布,酶联免疫吸附试验(ELISA)法检测 DN 患者和 HC 血浆中 IL-1 和 IL-1Ra 浓度。

结果

DN 组与 HC 组 IL-1(-511C/T)基因型和等位基因频率分布存在显著差异( < 0.05),DN 患者 T 基因型频率高于 HC 组(OR=2.84,95%CI:1.489-5.416)。IL-1(+3953C/T)和 IL-1Ra(+8006C/T)基因型和等位基因频率在两组间无显著差异( > 0.05)。DN 组血浆 IL-1 水平显著升高( < 0.01),IL-1Ra 浓度显著降低( < 0.05)。此外,IL-1(-511C/T)基因座变异体的血浆 IL-1 水平存在显著差异( < 0.05)。

结论

IL-1(-511C/T)基因多态性与中国桂北地区人群 DN 易感性显著相关,T 等位基因可能与 DN 的遗传易感性有关。
