Human Adenovirus species D (HAdV-D) was common human viral pathogen especially in eye infection, consists of several types of which HAdV-D8, -D19 and -D37 were common in eye infection. This study includes detection of HAdV-D types implicated in conjunctivitis based on L2 (Penton protein) gene similarity.
Conjunctival swabs were collected from Keratoconjunctivitis patients as eye infection related to adenovirus. Viral nucleic acids were extracted and specific primer pairs for HAdV-D L2 gene (encoding for penton base protein) was used to amplify the target gene and only positive samples were sent to sequencing.
The results revealed that only 6 samples give positive results for L2 gene amplification and then sent for sequencing for L2 (penton protein) gene-based typing. The results show that 4 local isolates (S1, S2, S3, S6) were similar to HAdV-D8 and 2 local isolates (S4, S5) were similar to HAdV-D20. Also the results display that the HAdV-37, prominent HAdV-D type of human eye infection, may be variant of HAdV-D20 due to that six variation were seen in S4and S5 local isolates nucleotide sequence in relation to HAdV-D37: T>C at position 14364, A>C at position 14411, T>C at position 14427, C>A at position 14448, G>A at position 14540 and T>C at position 14617, leading to only 2 amino acid change in resulted penton protein: T (Threonine) instead of K (Lysine) at position 204 and N (Asparagine) instead of D (Aspartic acid) at position 247.
The current study concludes the possibility of implication of HAdV-D20 in eye infections especially conjunctivitis.
人腺病毒D种(HAdV-D)是常见的人类病毒病原体,尤其在眼部感染中常见,它由几种类型组成,其中HAdV-D8、-D19和-D37在眼部感染中较为常见。本研究基于L2(五聚体蛋白)基因相似性检测与结膜炎相关的HAdV-D类型。
从腺病毒相关眼部感染的角结膜炎患者中采集结膜拭子。提取病毒核酸,使用针对HAdV-D L2基因(编码五聚体基底蛋白)的特异性引物对扩增目标基因,仅将阳性样本送去测序。
结果显示,仅6个样本L2基因扩增呈阳性,随后送去进行基于L2(五聚体蛋白)基因的分型测序。结果表明,4个本地分离株(S1、S2、S3、S6)与HAdV-D8相似,2个本地分离株(S4、S5)与HAdV-D20相似。此外,结果还显示,人眼部感染中常见的HAdV-37可能是HAdV-D20的变体,因为在S4和S5本地分离株的核苷酸序列中,与HAdV-D37相比有6处变异:第14364位T>C、第14411位A>C、第14427位T>C、第14448位C>A、第14540位G>A以及第14617位T>C,导致所产生的五聚体蛋白仅发生2个氨基酸变化:第204位由K(赖氨酸)变为T(苏氨酸),第247位由D(天冬氨酸)变为N(天冬酰胺)。
本研究得出HAdV-D20可能与眼部感染尤其是结膜炎有关的结论。
