Abstract

OBJECTIVE

To explore the effect of electroacupuncture (EA) on rats with chronic fallopian tube inflammation and its potential mechanisms.

METHODS

Thirty-six female Sprague-Dawley rats were divided into Control, Model and EA groups. The pathological morphology of the fallopian tubes was observed by hematoxylin-eosin (HE) and Masson staining. The results of transforming growth factor-β1 (TGF-β1), P38 mitogen-activated protein kinase (MAPK), phosphorylation (p)-p38MAPK in rat oviduct tissues were detected by immunohistochemistry. Results of P38MAPK, p-P38MAPK and TGF-β1 in rat oviduct tissues were detected by immunofluorescence. The expression level of p38MAPK, p-P38MAPK, TGF-β1 protein in rats was detected by Western blot. Quantitative real-time polymerase chain reaction (RT-qPCR) was used to detect mRNA expression levels of TGF-β1.

RESULTS

It found that collagen fibers counts decreased significantly in EA group compared to Model group. The phosphorylation of P38MAPK in EA group was significantly reduced compared to Model group. The serum TGF-β1 expressions in EA group increased decreased significantly.

CONCLUSION

Electroacupuncture was able to attenuate chronic salpingitis through down-regulating TGF-β1/MAPK signaling pathway.

目的

探讨电针对慢性输卵管炎模型大鼠的作用及其可能机制。

方法

将 36 只雌性 Sprague-Dawley 大鼠随机分为对照组、模型组和电针组。采用苏木精-伊红(HE)和 Masson 染色观察输卵管的病理形态学变化。免疫组织化学法检测大鼠输卵管组织中转化生长因子-β1(TGF-β1)、丝裂原活化蛋白激酶(MAPK)p38、磷酸化(p)-p38MAPK 的表达。免疫荧光法检测大鼠输卵管组织中 P38MAPK、p-P38MAPK 和 TGF-β1 的表达。Western blot 检测大鼠输卵管组织中 p38MAPK、p-P38MAPK、TGF-β1 蛋白的表达水平。采用实时定量聚合酶链反应(RT-qPCR)检测 TGF-β1mRNA 表达水平。

结果

与模型组比较,电针组大鼠输卵管组织中胶原纤维计数明显减少,p-P38MAPK 表达明显减少,血清 TGF-β1 表达明显降低。

结论

电针对慢性输卵管炎具有治疗作用,其机制可能与下调 TGF-β1/MAPK 信号通路有关。
