To establish a graphite furnace atomic absorption spectrometry method for the determination of trace gallium in whole blood. From January to May 2021, the five factors of ashing temperature, ashing time, atomization temperature, atomization time and matrix modifier concentration in the determination of gallium in whole blood by graphite furnace atomic absorption spectrometry were optimized by using L(16) (4(5)) orthogonal test design. At the same time, within-run, between-run, spiking recovery test and other methodological indicators were tested. Under the optimized detection conditions, the linear range of determination of gallium in whole blood by graphite furnace atomic absorption spectrometry was 0.29-100.00 μg/L (=0.9991) . The within-run and between-run relative standard deviations () of repetitive measurement at 10.0, 50.0, 80.0 μg/L concentration levels were 2.3%-4.4% and 1.5%-3.6%, the recovery rate of spiking was 98.1%-103.8%, and the detection limit of the method was 0.13 μg/L. Graphite furnace atomic absorption spectrometry for the determination of trace gallium in whole blood is easy to operate, has a wide linear range, low detection limit, accurate and reliable results, which is suitable for occupational health examinations and the determination of acute gallium poisoning.
建立石墨炉原子吸收光谱法测定全血中痕量镓的方法。2021年1月至5月,采用L(16)(4(5))正交试验设计优化石墨炉原子吸收光谱法测定全血中镓时的灰化温度、灰化时间、原子化温度、原子化时间和基体改进剂浓度这五个因素。同时,进行批内、批间、加标回收试验等方法学指标检测。在优化的检测条件下,石墨炉原子吸收光谱法测定全血中镓的线性范围为0.29~100.00μg/L(r=0.9991)。在10.0、50.0、80.0μg/L浓度水平重复测定的批内和批间相对标准偏差(RSD)分别为2.3%~4.4%和1.5%~3.6%,加标回收率为98.1%~103.8%,方法检出限为0.13μg/L。石墨炉原子吸收光谱法测定全血中痕量镓操作简便,线性范围宽,检出限低,结果准确可靠,适用于职业健康检查及急性镓中毒的测定。
