Abstract

INTRODUCTION

is an Iranian herbal medicine with various effects on the human cells. The aim of this study was to investigate the effects of the aqueous extract of (AEAM) on the proliferation and differentiation of mesenchymal stem cells (MSCs).

METHODS

In this study, bone marrow-MSCs (BM-MSCs) were obtained from Wister rat bone morrow and then cultured in Dulbecco's modified Eagle's medium /Nutrient Ham's Mixture F-12 (DMEM/F12) media. Then, the isolated MSCs were cultured in either osteocyte or adipocyte differentiation media containing 0.2 or 2 mg/mL AEAM and assessed using specific staining method.

RESULTS

The isolated BM-MSCs exhibited fibroblast-like morphology and were positive for CD73, and CD90, while negative for CD34 and CD45. AEAM significantly increased self-renewal of BM-MSCs at low dose (0.2 mg/ml, P= 0.001) and increased the pool stem cells in both osteocyte and adipocyte differentiation media.

CONCLUSION

AEAM at low doses may be used in cases where there is a need for large number of stem cells, via increased numbers of MSCs, and help tissue repair and immunomodulation.

引言

是一种对人体细胞有多种作用的伊朗草药。本研究的目的是调查(某种草药名称未给出,推测为前文提到的草药)水提取物(AEAM)对间充质干细胞(MSCs)增殖和分化的影响。

方法

在本研究中,从Wister大鼠骨髓中获取骨髓间充质干细胞(BM - MSCs),然后在杜氏改良 Eagle 培养基/营养混合 Ham's F - 12(DMEM/F12)培养基中培养。接着,将分离出的MSCs在含有0.2或2mg/mL AEAM的成骨细胞或脂肪细胞分化培养基中培养,并使用特定染色方法进行评估。

结果

分离出的BM - MSCs呈现成纤维细胞样形态,CD73和CD90呈阳性,而CD34和CD45呈阴性。低剂量(0.2mg/ml,P = 0.001)的AEAM显著增加了BM - MSCs的自我更新能力,并增加了成骨细胞和脂肪细胞分化培养基中的干细胞库。

结论

低剂量的AEAM可用于需要大量干细胞的情况,通过增加MSCs数量,有助于组织修复和免疫调节。
