Abstract

As an important endogenous gasotransmitter, hydrogen sulfide (HS) has been implicated with a variety of biological processes and has attracted more and more attention for its key role in a wide range of pathological processes. However, lacking tools for HS-specific in situ detection, the changes of endogenous HS levels in the pathological progression of diseases are still unclear. In this work, a turn-on fluorescent probe (BF-DBS) has been designed and synthesized by two-step reactions using 4-diethylaminosalicylaldehyde and 1,4-dimethylpyridinium iodide as raw materials. Probe BF-DBS displays high selectivity and sensitivity to HS with a large Stokes shift and good anti-interference ability. The practical application of probe BF-DBS to detect endogenous HS was evaluated in living HeLa cells.

作为一种重要的内源性气体信号分子,硫化氢 (HS) 参与了多种生物学过程,并因其在广泛的病理过程中的关键作用而受到越来越多的关注。然而,由于缺乏 HS 特异性的原位检测工具,疾病病理进展过程中内源性 HS 水平的变化仍不清楚。在这项工作中,我们设计并合成了一种基于双醛缩苯胺和 1,4-二甲基吡啶𬭩碘盐的两步反应的荧光探针 (BF-DBS)。探针 BF-DBS 对 HS 具有高选择性和灵敏度,Stokes 位移大,抗干扰能力好。探针 BF-DBS 对活 HeLa 细胞内源性 HS 的检测的实际应用进行了评价。
