A total of 24,000 healthy 1-day-old Arbor Acres broilers with similar initial weights were used in this study and fed a basal diet supplemented with 0, 400 and 800 mg/kg isoleucine (Ile), denoted CON, ILE400 and ILE800, respectively. Results revealed that the final body weight, average daily weight gain, and eviscerated carcass rate, of broiler chickens in the ILE400 group were significantly higher than in other groups ( < 0.05). In addition, the ILE400 and ILE800 groups had a lower feed conversion rate and a higher survival rate and breast muscle rate ( < 0.05), while the abdominal fat rate was significantly lower than the CON group ( < 0.05). There were significantly lower serum concentrations of UREA, glucose (GLU) and total cholesterol (TCHO) in the ILE400 and ILE800 groups than in the CON group ( < 0.05); glutathione peroxidase (GSH-Px) activity was significantly higher in the ILE400 group than in the other groups, and tumor necrosis factor-alpha (TNF-α) concentration was considerably lower than in other groups ( < 0.05). Moreover, interleukin (IL)-10 concentration in the ILE800 group was significantly higher than in the other groups ( < 0.05). The ILE400 group significantly down-regulated the mRNA expressions of fatty-acid synthase () and solid alcohol regulatory element binding protein 1c (), and significantly up-regulated the mRNA expressions of adipose triglyceride lipase (), hormone-sensitive lipase (), lipoprotein lipase () and sirtuin1 () ( < 0.05). The ILE400 group had significantly higher intestinal villus height than the CON and ILE800 groups, while the ILE800 group had significantly lower intestinal villus height/crypt depth ( < 0.05). Furthermore, high-throughput sequencing showed that the Shannon index, and , and abundances were significantly higher in the ILE400 group than in the CON group ( < 0.05). Interestingly, the ILE800 group reduced the Simpson index, phylum and abundances (including genera , , and ), and increased and (including genera , , and ) ( < 0.05). Overall, our study showed that adding 400 mg/kg Ile to the diet (diets total Ile levels at 1.01%, 0.90% and 0.87% during the starter, grower and finisher phases, respectively) increased production performance and improved the health status in broiler chickens.
本研究共选用24000只初始体重相近的1日龄健康艾维茵肉鸡,饲喂基础日粮,并分别添加0、400和800 mg/kg异亮氨酸(Ile),分别记为CON、ILE400和ILE800。结果显示,ILE400组肉鸡的末体重、平均日增重和屠体率显著高于其他组(P<0.05)。此外,ILE400和ILE800组的料重比更低,成活率和胸肌率更高(P<0.05),而腹脂率显著低于CON组(P<0.05)。ILE400和ILE800组血清尿素、葡萄糖(GLU)和总胆固醇(TCHO)浓度显著低于CON组(P<0.05);ILE400组谷胱甘肽过氧化物酶(GSH-Px)活性显著高于其他组,肿瘤坏死因子-α(TNF-α)浓度显著低于其他组(P<0.05)。此外,ILE800组白细胞介素(IL)-10浓度显著高于其他组(P<0.05)。ILE400组显著下调脂肪酸合酶(FAS)和固醇调节元件结合蛋白1c(SREBP-1c)的mRNA表达,显著上调脂肪甘油三酯脂肪酶(ATGL)、激素敏感性脂肪酶(HSL)、脂蛋白脂肪酶(LPL)和沉默信息调节因子1(SIRT1)的mRNA表达(P<0.05)。ILE400组肠绒毛高度显著高于CON组和ILE800组,而ILE800组肠绒毛高度/隐窝深度显著更低(P<0.05)。此外,高通量测序显示,ILE400组的香农指数、Chao1指数、Ace指数和OTU丰度显著高于CON组(P<0.05)。有趣的是,ILE800组降低了辛普森指数、门水平和科水平丰度(包括属水平的Prevotella、Turicibacter、Erysipelotrichaceae_UCG-003和Ruminococcus_1),增加了纲水平和目水平丰度(包括属水平的Lactobacillus、Turicibacter、Erysipelotrichaceae_UCG-003和Ruminococcus_1)(P<0.05)。总体而言,我们的研究表明,在日粮中添加400 mg/kg异亮氨酸(分别使雏鸡、中鸡和大鸡阶段日粮总异亮氨酸水平达到1.01%、0.90%和0.87%)可提高肉鸡的生产性能并改善其健康状况。
