DNA methylation is a stable and heritable epigenetic mark, and it plays an important role in regulation of gene expression and transposon silencing. Here we developed a CRISPR/dCpf1-based targeted demethylation system using the catalytic domain of the human demethylase TEN-ELEVEN TRANSLOCATION1 (TET1cd) and a SunTag system. The SunTag-dCpf1-TET1cd system is able to achieve targeted DNA demethylation and up-regulate gene expression when guided to the or loci in . Our study provides tools for targeted removal of DNA cytosine methylation, and activation of protein-coding genes or transposons expression.
DNA甲基化是一种稳定且可遗传的表观遗传标记,它在基因表达调控和转座子沉默中发挥着重要作用。在此,我们利用人类去甲基化酶TEN-ELEVEN TRANSLOCATION1(TET1cd)的催化结构域和SunTag系统,开发了一种基于CRISPR/dCpf1的靶向去甲基化系统。当被引导至人细胞的特定基因座时,SunTag-dCpf1-TET1cd系统能够实现靶向DNA去甲基化并上调基因表达。我们的研究为靶向去除DNA胞嘧啶甲基化以及激活蛋白质编码基因或转座子表达提供了工具。
