Among the most important transcription factors in plants, the v-myb avian myeloblastosis viral oncogene homolog () regulates the expression network of response genes under stresses such as fungal infection. In China, the canker disease threatens the survival of , an ancestor of cultivated apples. Using the genome, we identified 457 and 128 genes that were randomly distributed across 17 chromosomes. Based on protein sequence and structure, the genes were phylogenetically divided into 29 categories, and 26 conserved motifs were identified. We further predicted -elements in the 2000-kb promoter region of s based on the genome. Transcriptome analysis of . under infection showed that 27 were significantly differentially expressed, indicating their key role in the response to infection. Using transient transformation, , , , , and , which were strongly induced by infection, were functionally identified. Among the five , and were both important in enhancing resistance to diseases, whereas inhibited resistance. Based on the results of this study, we gained a better understanding of the transcription factor family and laid the foundation for a future research program on disease prevention strategies in .
在植物中最重要的转录因子中,v-myb禽成髓细胞瘤病毒癌基因同源物()调节真菌感染等胁迫下响应基因的表达网络。在中国,溃疡病威胁着栽培苹果的祖先——山荆子的生存。利用山荆子基因组,我们鉴定出457个v-myb和128个相关基因,它们随机分布在17条染色体上。基于蛋白质序列和结构,v-myb基因在系统发育上被分为29类,并鉴定出26个保守基序。我们进一步基于基因组预测了山荆子2000 kb启动子区域中的顺式作用元件。对山荆子感染溃疡病菌后的转录组分析表明,27个v-myb基因显著差异表达,表明它们在响应溃疡病菌感染中起关键作用。通过瞬时转化,鉴定出了受溃疡病菌感染强烈诱导的MdbHLH3、MdbHLH13、MdbHLH17、MdbHLH100和MdbHLH101的功能。在这五个MdbHLH中,MdbHLH3和MdbHLH101在增强抗病性方面都很重要,而MdbHLH17抑制抗病性。基于本研究结果,我们对v-myb转录因子家族有了更好的理解,并为未来山荆子疾病预防策略的研究计划奠定了基础。
